Abstract
introduction the plant periwinkle (madagascan periwinkle) plant (catharanthus roseus l.), an ornamental and medicinal herb, is indigenous to madagascar and tropical regions. it is valued for its high phenolic compounds content as well as significant commercial medicinal alkaloids, including ajmalicine, vincristine, and vinblastine. c. roseus (vinca) exhibits prominent therapeutic properties, including anti-cancer effect, antihypertensive activity, wound-healing effects, and blood-purifying potentials. it is also used to treat human skin disorders such as eczema, as well as urinary tract infections and bronchial inflammation. the cultivation of c. roseus is subject to various environmental stresses, particularly drought, which profoundly impacts its biochemical profiles and the production of secondary metabolites. the application of certain elicitors, notably the tryptophan amino acid, has been shown to alleviate the detrimental effects of drought stress and enhance the overall quality of agricultural products, thereby improving plant performance. moreover, tryptophan is recognized as a precursor in the alkaloid biosynthesis pathway. consequently, it is plausible that the combination of drought stress with foliar application of tryptophan may elevate the levels of plant metabolites, particularly vinblastine and vincristine alkaloids. materials and methods this study was conducted using potted c. roseus plants grown in 1:1:1 ratio of farm soil, decomposed animal manure, and sand. the experiment was conducted in a 6×3 factorial experiment arranged in a completely randomized design with three replications. the treatments consisted of six tryptophan treatments (0 (control), 50, 100, 150, 200, and 250 mg/l) and three drought stress level (100%, 70%, and 40% of field capacity). the experiment was carried out in 2023 at khuzestan university of agricultural sciences and natural resources under greenhouse conditions. foliar sprays of tryptophan solutions were applied at two phenological stages: eight-leaf vegetative phase and floral initiation. morphological traits such as root volume, dry weight, and length were measured. plus, the effects of different treatment on str and tdc gene expression patterns were evaluated. morphological data were analyzed using sas software (version 9.4), while relative gene expression was quantified with rest software. the means were compared using duncan's multiple range test at p<0.05 significance level. results and discussion the interaction effect of drought stress and foliar application of the tryptophan amino acid on root volume and dry weight traits was significant (p<0.05); however, no significant difference was observed for root length. results of present study demonstrated contrasting responses to drought and tryptophan treatments. under increasing drought stress, root length significantly (p<0.05) increased, while root volume and dry weight decreased significantly (p<0.05) compared to controls. tryptophan application dose-dependently enhanced all root morphological parameters, with optimal concentration, producing maximal improvements (p<0.05). relative gene expression analysis showed that the combined treatment of 250 mg/l tryptophan under severe drought stress (40% fc) significantly up-regulated tdc and str genes expression of c. roseus leaf tissues after 24 hours, followed by a decreasing trend thereafter. the lowest relative expression levels of tdc and str genes were recorded at 72 and 168 hours, with values of -3.69 and -1.18, respectively. given the increased expression of tdc and str genes due to foliar application compared to the control treatment, it is anticipated that the end products of the tias pathway, including the alkaloids ajmalicine, vinblastine, and vincristine, may also increase. the coordinated increase in root morphological traits, i.e. root volume, root weight and root length as well as the upregulation of tia pathway genes, i.e. tdc, str strongly suggests that foliar tryptophan application represents a viable physiological strategy to enhance vincristine and vinblastine production in c. roseus. this dual effect can likely be explained by increased precursor availability for alkaloid biosynthesis, and drought-stress-mediated activation of secondary metabolic pathways. conclusions the findings of present study suggest that foliar application of the tryptophan amino acid, particularly at concentrations of 200 and 250 mg/l, resulted in increased root length and dry weight. tryptophan amino acid treatment not only induced tolerance in drought-stressed plants but also improved the root system, leading to enhanced dry weight. foliar application of tryptophan significantly upregulated main genes associated with terpenoid indole alkaloid (tia) biosynthetic pathway, with treated plants exhibiting a higher expression level of tdc and str compared to controls (p<0.05). this enhanced gene expression correlated with increased alkaloid production potential.
