چکیده
|
تنش فلزی یکی از مشکلات جدی است که تولید محصولات گیاهی را در سراسر جهان محدود می کند. تنش فلزی اثرات متعددی از جمله تغییرات فنوتیپی، آنزیمی و بیان ژنی در گیاهان دارد. گندم یک محصول راهبردی در تغذیه انسان بهشمار میرود و افزایش روزافزون جمعیت جهانی مستلزم تولید بیشتر محصولات کشاورزی است. با توسعه سریع صنعت و کشاورزی مدرن، آلودگی فلزات سنگین در نتیجه فعالیتهای انسانی به یکی از چالشهای مهم جهانی تبدیل شده است. بر همین اساس در این پژوهش آزمایشی بهصورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی در سه تکرار انجام شد. در این مطالعه اثر فاکتورهای آزمایش شامل نیتراتنقره در سه سطح (0، 1، 2 میلی مولار) بهعنوان عامل ایجاد تنش اکسیداتیو و دبکو (dabco) بهعنوان آنتیاکسیدان در سه سطح (0، 10، 20 میلی مولار) روی رقم فلات و تجن گندم مورد بررسی قرار گرفت. صفات بیوشیمیایی مورد ارزیابی شامل شاخص سطح اکسیداسیونی سلول و محتوای کلروفیل بودند، همچنین ارزیابی بیان ژن های گلوکوناز و متالوتیونین به روش real time qrt-pcr انجام شد. نتایج بررسی صفات میزان کلروفیل a و b نشان داد که با افزایش غلظت نیتراتنقره، میزان این صفات کاهش قابل توجهی پیدا کردند. اعمال پیشتیمار دبکو قبل از محلول پاشی نیتراتنقره منجر به افزایش در میزان این صفات به ویژه در غلظت 1 میلیمولار نیتراتنقره گردید. در شرایط تنش، بیان ژن متالوتیونین و گلوکاناز در سطوح پایین تر تنش افزایش یافت، اما با افزایش سطحوح بالاتر نیتراتنقره، بیان این ژن ها کاهش یافتند. در هر دو رقم، سطح بالای بیان ژن های متالوتیونین و گلوکاناز در تیمار 1 میلیمولار نیتراتنقره بدون ترکیب با دبکو و کاهش بیان ژن در نیتراتنقره 2 میلیمولار مشاهده شد. اگرچه در این تحقیق بهطور خاص تاثیر ترکیب دبکو بررسی نشد، اما اثر آنتیاکسیدانها در کاهش تنش اکسیداتیو در گیاهان بهطور عمومی شناخته شده است. با توجه به تاثیر مثبت دبکو بر کاهش اثرات تنش فلزی، پیشنهاد میشود در کشتهای گندم از این ترکیب بهعنوان یک روش مدیریتی بهرهبرداری شود. همچنین بررسی دقیقتر روی دیگر ژنهای مرتبط با پاسخ به تنش اکسیداتیو میتواند به ما در درک بهتر سازوکارهای مقاومت گیاهان کمک کند. مطالعه شرایط بهینه برای استفاده از نیترات نقره و دبکو میتواند به افزایش تولید و بهبود کیفیت محصولات کشاورزی کمک کند.

Abstract
|
introduction metal stress is a major challenge affecting agricultural production, given the increasing global population and the critical need for food. heavy metal pollution remains a serious threat to production and quality of wheat (triticum aestivum l.), as a major crop for human nutrition. understanding the effects of metal stress on plants and their response mechanisms is crucial for optimizing agriculture. metal stress in plants is commonly induced by heavy metals such as silver, cadmium, lead, and copper. such heavy metals adversely affect plant growth when present in soil, water, or air. heavy metal stress can lead to various changes in plant metabolism, including, enzymatic stress, and gene expression alterations. due to high nutritional requirements and sensitivity to environmental changes, wheat is particularly susceptible to heavy metals-induced stress. understating the impacts of heavy metals on wheat is crucial for enhancing wheat quality and quantity. developing resistant varieties and effective agronomic management techniques are crucial to combat wheat production challenges worldwide. this study aimed to explore the impact of silver nitrate (agno₃) as an oxidative stress agent and dabco as an antioxidant on different wheat varieties (flat and tajan) and the expression of specific genes, metallothionein and glucanase. materials and methods a factorial experiment was conducted in a randomized complete block design with three replications. the experimental factors included silver nitrate at three levels (0, 1, 2 mm) as an oxidative stress factor and dabco as an antioxidant at three levels (0, 10, 20 mm) on wheat cultivars falat and tajan. biochemical traits assessed included the cell oxidation level index and chlorophyll content, and the expression of gluconase and metallothionein genes was evaluated using real time qrt-pcr. total rna extraction was conducted using bioflex p-biosol extraction buffer. the quality of the extracted rna was assessed by on 1.5% agarose gel electrophoresis. subsequently, cdna was synthesized according to fermentase manufacturer instructors. the quality of cdna was then assessed by pcr using specific primers for the housekeeping gene gapdh. in order to analyze the expression patterns of the gluconase and metallothionein genes, real time qrt-pcr was performed utilizing cybergreen i dye. real-time qrt-pcr analysis was performed using the cyberbioparse kit at gorgan university of agricultural sciences and natural resources in iran and on an iq 5 device made in the united states.results and discussion the results of present study demonstrated that under silver nitrate stress, both cultivars showed the highest level of gene expression at 1 mm silver nitrate treatment without debco treatment, followed by significant downregulation at 2 mm silver nitrate treatment. these results may imply that wheat is sensitive to a high oxidative stress levels after silver nitrate treatment. increased expression of metallothionein and glucanase at lower concentrations may serve as a protective response to oxidative stress, neutralizing oxidant effects and safeguarding cells from metal stress damage. additionally, at 1 mm silver nitrate treatment, the application of dabco as an antioxidant, positively influenced associated genes expression, possibly leading to reduced oxidative damage. conclusion the present study demonstrates that silver nitrate, as an oxidative stress inducer, has significant effects on the expression of genes associated with heavy metal stress in wheat. scientific reports indicate that silver nitrate increases the expression of metallothionein and glucanase genes, where they play a key role in plants' response to environmental stresses. metallothioneins, as metal-binding proteins, are crucial in neutralizing the toxic effects of heavy metals and reducing oxidative stress. additionally, glucanases contribute to the breakdown of carbohydrates and may also impact metabolic changes in response to metal stress conditions. the observed gene expression variations at different levels of silver nitrate and types of stresses suggest that wheat can adapt to stress in order to regulate corresponding genes to cope with environmental challenges. these adaptations may lead to genetic changes and natural selection in the long term, potentially enabling plants to survive longer under harsh conditions. considering the global significance of wheat as a strategic food crop, the results of this study underscore the necessity for further research in the field of metal stress management. one potential future objective could involve identifying wheat cultivars better equipped to handle metal stresses. these cultivars could be developed through genetic engineering or plant breeding programs to enhance yield and quality of agricultural products. ultimately, it is crucial for agricultural scientists and researchers to collaborate in order to provide effective solutions to mitigate the negative impacts of metal stress and contribute to sustainable food security in the future. this research could serve as a foundation for developing solutions that not only ensure plant health but also promote the health of agricultural ecosystems.
