Phylogenetic analysis of Pasteurella multocida isolates recovered from fowl cholera outbreaks in geographically related poultry flocks

Six pasteurella (p.) multocida isolates were recovered out of 43 liver samples collected from geographically related commercial poultry flocks in and around faisalabad,pakistan. the isolates were identified based on conventional biochemical tests and analyticalprofile index (api 20-e) profiles. the isolates were quite similar in their biochemical profiles whereas api 20-e profiles indicated 1.4 to 13.9 % variations. molecular characterization was done by random amplified polymorphic dna polymerase chain reaction (rapd-pcr)through amplification of genomic dna to find out genetic variations. phylogenetic analysis,based on similarity coefficient which was generated through amplification of p. multocida genomic dna,indicated 22.7 to 41.4% genetic variations. in addition,obtained phylogenetic tree was a single lineage with branched pattern which showed a genetic linkage among isolates of p. multocida. based on the results,it was concluded that rapd-pcr amplification could be a useful tool to investigate genetic variations among various bacterial isolates from commercial poultry flock,even in geographically related areas.
