بررسی ارتباط پلی­مورفیسم پروموتر Mmp-3 و غلظت سرمی آن با نتیجه لقاح آزمایشگاهی و انتقال جنین

هدف از این تحقیق بررسی پلی مورفیسم ژن mmp3 و بیان آن در سرم خون در زنان ناباروری است که لقاح مصنوعی و انتقال جنین (in vitro fertilization and embryo transfer) انجام داده اند. در این مطالعه100 زن نابارور با نتیجه لقاح مصنوعی ناموفق (ivf) و 100 زن بارور با نتیجه لقاح مصنوعی و حاملگی کلینیکی موفق (ivf+) مورد بررسی قرار گرفتند. پلی‌مورفیسم ژنی و غلظت سرمی mmp3 به ترتیب توسط واکنش زنجیره‌ای پلیمراز (armspcr) و روش الایزا (elisa) مورد بررسی قرار گرفت. نتایج به دست آمده بیانگر عدم وجود ارتباط بین فراوانی آللی و ژنوتیپی ژن mmp3 در دو گروه مورد مطالعه است. همچنین نشان داده شد که کاهش معنی‌داری در غلظت mmp3 بین دو گروه وجود دارد (0.000002=p). نتایج نشان داد میزان سرمی mmp3 در ژنوتیپ های aa، ac و cc در گروه ivf به ترتیب مقادیر 86، 65.33 و 33 ng/ml است. به طور کلی نتیجه‌گیری می‌شود که ارتباط معناداری بین پلی‌مورفیسم پروموتر ژن mmp3 با دو گروه ivf+ و ivf وجود ندارد، در حالی که کاهش معناداری در سطح پروتئین سرمی mmp3 در گروه ivf در مقایسه با گروه ivf+ وجود دارد. همچنین نشان دادیم که ژنوتیپ cc با کاهش غلظت سرمی mmp3 در ارتباط است و ممکن است در نتیجه لقاح مصنوعی و انتقال جنین نقش داشته باشد.

Analyzing the association of MMP-3 promoter polymorphism and its serum concentration with the outcome of in vitro fertilization and embryo transfer

This study aimed to investigate the polymorphism of matrix metalloproteinase 3 (MMP3) gene and its expression in the serum of infertile female patients received in vitro fertilization and embryo transfer (IVFET). To do so, 100 women with unsuccessful IVFET (IVF -) and 100 women with successful IVFET procedure and clinical pregnancy (IVF+) were included. Genetic polymorphism and serum concentration of MMP3 were investigated by ARMSPCR and ELISA, respectively. The results showed no significant association between MMP3 gene polymorphism and IVFET outcome among the two groups studied. However, a significant decrease in the concentration of MMP3 serum in the IVF - group was observed in comparison with the IVF+ group (P=0.000002). Moreover, we showed that the serum MMP3 levels in CC, AC and AA genotypes in the IVF - group were 33, 65.33 and 86 ng/ml, respectively. In conclusion, while there is no significant difference between MMP3 promoter polymorphism and IVFET outcome between the IVF+ and IVF groups, a significant decrease in MMP3 serum levels in IVF group was seen as compared with the IVF+ group. It could be also suggested that the CC genotype is associated with a decreased level of MMP3 serum concentration and may be associated with IVFET failure.
