   متیلاسیون پروموتر ژنهای P14, P15 و P16 در مبتلایان ایرانی سرطان بافت سنگفرشی مری

چکیده    مطالعات نشان داده است که متیلاسیون نابه جای ژنهای سرکوبگرتومور باعث خاموش شدن آنها و درنتیجه کاهش بیان آنها شده و زمینه برای ایجاد سرطان مهیا می شود. هدف از این مطالعه، بررسی متیلاسیون سه ژن سرکوبگرتومورp14,p15و p16 در مبتلایان به سرطان بافت سنگفرشی مری (escc) و ارتباط آن با خصوصیات دموگرافی و پاتولوژیکی بیماران می باشد. به این منظور از 44نفر بیمار escc غیرخویشاوند، 44عدد نمونه بافت توموری و 19عدد نمونه سالم(مجاور تومور)تهیه و مورد استخراجdna وrnaقرار گرفت. سپس باپرایمر اختصاصی حالتهای متیله وغیرمتیله این ژنها،واکنش زنجیره ای پلیمراز ویژه متیلاسیون(mspcr)انجام شد. بیان این ژنها نیزباrtpcrبررسی گردید. نتایج نشان داد درمیان نمونه های توموری، درصد متیلاسیون برای ژنهایp14،p15وp16 به ترتیب عبارتست از 53%،9%و27% و در هیچ یک از نمونه های نرمال(مجاور تومور)متیلاسیونی مشاهده نشدونیزمیزان بیان این ژنها با میزان متیلاسیون نسبت عکس داشت(p<0.05). فراوانی متیلاسیون پروموتر ژنهای p15وp14در نمونه های توموری scce به ترتیب 41%، 9% وفراوانی وجود متیلاسیون همزمان در ژنهای p15+p16 و p14+p15 به ترتیب 6/13% و 5/4% بود درحالیکه فراوانی متیلاسیون در ژن های p14+p16، 9% مشاهده شد (p=0.0036).بدین معنا که در تمامی بیماران متیله برای ژن p14،متیلاسیون p16 مشاهده شداما در بیشتر بیمارانی کهp15متیله بود، ژن های p14ویا p16 متیلاسیون نشان نداد.مقایسه این نتایج با سایر یافته های مشابه در جهان،نشان میدهد وضعیت متیلاسیون ژن های p15،p14،p16در جمعیت ایرانی مورد بررسی ‌در محدوده گزارشاتی از شمال چین قرار دارد. بعبارتی احتمالاً مکانیسم‌های مولکولی و عوامل اتیولوژیک مشابهی میتوانند در بروز سرطان escc در این دو جمعیت (ایران و چین) نقش داشته باشند.

   Investigation of the aberrant DNA methylation of p14, p15, p16 genes in patients with ESCC in Iran

Abstract    Studies have shown that aberrant methylation of the tumor suppressor genes could be lead to silencing and reducing of their gene expression, consequently occurring the cancer.The aim of this study was investigation of the aberrantDNAmethylation of p14,p15,p16 genes in Iranian patients with Esophageal Squamous cell carcinoma(ESCC) and its association with demographic and pathological characteristics. Therefore we extracted the DNAs and RNAs of44 tumor tissues and 19 normal tissues from 44 unrelated Iranian patients with ESCC. The methylation specific PCR(MSPCR)was performed for both methylated and unmethylated status of these genes and the expression of these genes have done with RTPCR.The results showed the 53%,9% and 27% methylation for the p14,p15 and P16 genes respectively in the tumor tissues and no methylation frequency in none of the normal sample. Regarding the gene expression, there was an inverse relationship between methylation status and gene expression(p<0.05).The frequency of methylation of p15 and p14 in tumor samples was 41%,9% respectively and abundance of methylation the p15+p16 and p14+p15, was 6/13%, and 5/4%, respectively, while the frequency of methylation in p14+p16 genes was observed 9% (P = 0.0036). It means that all patients which are methylated for the p14 had the methylation in p16 too while in most patients with p15 methylated, the p14 or p16 methylation was not found. Comparing these results with other reports shows a similarity between the Iranian population and Chinese population (North of China) maybe because of a similar molecular mechanisms and etiology of ESCC cancer in these populations.
