   تاثیر گاما آمینو بوتیریک اسید بر شاخص‌های فیزیولوژیکی و بیوشیمیایی برگ دو رقم انگور (vitis vinifera l.) تحت تنش سرما

چکیده    در این پژوهش، اثر کاربرد برگی گاما آمینو بوتیریک اسید (گابا؛ صفر، 1.5 و 3 میلی مولار) بر محتوای آبسیزیک اسید، قندهای محلول، پلی آمین ها، فعالیت برخی آنزیم‌های آنتی‌اُکسیدان و شاخص‌ پایداری غشاء برگ دو رقم انگور (خلیلی و پرلت) تحت دو تیمار دمایی (ºc2 و ºc24) به صورت فاکتوریل (2×3) در قالب طرح کاملاً تصادفی با سه تکرار در شرایط گلخانه بررسی شد. تاک‌ها در مرحله 15 برگی با غلظت‌های مختلف گابا محلول‌پاشی شدند. بعد از 24 ساعت، نیمی از تاک‌های تیمارشده با گابا به همراه گیاهان شاهد به مدت 12 ساعت تحت دمای ºc2 (شرایط تنش سرما) قرار گرفتند و بقیه تاک‌ها (تیمارشده با گابا و شاهد) در دمای ºc24 (شرایط عادی) نگهدای و بعد 12 ساعت نمونه برداری شدند. کاربرد گابا در شرایط تنش سرما سبب حفظ کلروفیل و کاهش نشت یونی، مالون دی آلدئید و پراکسید هیدروژن و افزایش پروتئین، پرولین و آبسیزیک اسید نسبت به گیاهان شاهد در هر دو رقم شد. مقدار ساکارز، فروکتوز و گلوکز برگ در رقم خلیلی تیمار شده در مقایسه با رقم پرلت تحت تنش سرما بیشتر بود. همچنین کاربرد گابا منجر به افزایش مقدار پلی آمین‌های پوتریسین، اسپرمین و اسپرمیدین در دمای ºc2 شد. تحت تنش سرما، فعالیت آنزیم های سوپراکسید دیسموتاز و کاتالاز در غلظت 3 میلی مولار و آسکوربات پراکسیداز در غلظت 1.5 میلی مولار گابا بیشترین فعالیت آنزیمی را نسبت به گیاهان شاهد نشان دادند. گابا از طریق افزایش آبسیزیک اسید، قندهای محلول و سیستم آنتی‌اکسیدانی، باعث حفظ پایداری غشاء و تحمل به سرمای تاک‌ها شد. تاثیر گابا بر تحمل به سرمای رقم خلیلی بیشتر از رقم پرلت بود.

   the effect of gamma-aminobutyric acid on leaf physiological and biochemical indices of two grape (vitis vinifera l.) cultivars under cold stress

Abstract    in this research, the effect of foliar application of gamma-amino-butyric acid (gaba; 0, 1.5 and 3 mm) on abscisic acid, soluble sugars, polyamines, the activity of some antioxidant enzymes, and membrane stability index of two grape cultivars (khalili and perlette) was investigated under greenhouse condition with two temperature treatments (2ºc and 24ºc) factorially (2×3) based on a completely randomized design with three replications. the vines were sprayed with different concentrations of gaba at the 15-leaf stage. after 24 hours, half of the vines treated with gaba along with the control plants were exposed to 2ºc (cold stress condition) for 12 hours and the rest vines (treated with gaba and control) were kept at 24ºc (non-stress condition) and sampled after 12 hours. the application of gaba under cold stress conditions caused a preservation of chlorophyll and a decrease of ion leakage, malondialdehyde and hydrogen peroxide and increase of protein, proline and abscisic acid content compared to control plants in both cultivars. the amount of sucrose, fructose and glucose in the leaves was higher in the treated khalili cultivar compared to the perlette cultivar under cold stress. also, under cold stress, gaba spray led to an increase in the amount of polyamines of putrescin, spermine and spermidine. under cold stress, the activities of superoxide dismutase and catalase enzymes at a concentration of 3 mm and ascorbate-peroxidase at a concentration of 1.5 mm of gaba showed the highest value compared to the control plants. gaba treatment maintained membrane stability and increased cold tolerance of vines by increasing in abscisic acid content, soluble sugars and antioxidant system, the effect of gaba on the cold tolerance of khalili cultivar was greater than that of perlette cultivar.
