ارتباط چندشکلی vntr ژن nlrp3 با خطر ابتلا به سرطان معده

زمینه و هدف: سرطان معده یکی از سرطان‌های شایع در سراسرجهان از جمله ایران می‌باشد که مرگ ومیر بالایی دارد. یکی از عوامل موثر در ابتلا به سرطان معده، التهاب ناشی از افزایش عملکرد پروتئین cryopyrin است که در انسان توسط ژن nlrp3 کد می‌شود. افزایش بیان این ژن منجر به تولید سایتوکاین های il-1β و il-18 شده و التهاب را افزایش می­دهد. با توجه به نقش مهم ژن nlrp3، در این مطالعه نقش چندشکلی vntr در اینترون 4 ژن nlrp3 در استعداد ابتلا به سرطان معده بررسی شد.مواد و روش­ها: در این مطالعه چندشکلی vntr ژن nlrp3 در 100 فرد مبتلا به سرطان معده و 88 فرد سالم با تکنیکpcr  تعیین ژنوتیپ گردید. داده‌های به‌دست‌آمده با نرم‌افزار spss ν21 و آزمون رگرسیون لجستیک آنالیز شدند.یافته­ها: در این مطالعه 15 نوع ژنوتیپ مختلف و 7 نوع آلل در پلی‌مورفیسم vntr ژن nlrp3 شناسایی شد که آلل 12 تکرار و ژنوتیپ 12.12 بیشترین فراوانی را به خود اختصاص داد. همچنین برای اولین بار آلل 14 تکرار شناسایی گردید. آنالیز نتایج نشان داد که ژنوتیپ 12.12 در مقایسه با سایر ژنوتیپ خطر ابتلا به سرطان معده را افزایش می‌دهد ) 0.01 p value=، 0.76 - 0.14 ci=، 0.32(or=.نتیجه: به نظر می‌رسد ژنوتیپ 12.12 در اینترون 4 ژن nlrp3 خطر ابتلا به سرطان معده را افزایش می‌دهد.

background and aim: gastric cancer is one of the most common cancers in the world, including iran, which has a high mortality rate. one of the effective factors in the development of gastric cancer is the increase in inflammation caused by the increase in the function of the cryopyrin protein, which is encoded by the nlrp3 gene in humans. increased expression of this gene leads to the production of il-1β and il-18 cytokines and increases inflammation. considering the important role of nlrp3 gene, in this study, the role of vntr polymorphism in intron 4 of nlrp3 gene in susceptibility to gastric cancer was investigated.materials and methods: in this study, vntr polymorphism of nlrp3 gene was genotyped in 100 patients with gastric cancer and 88 healthy individuals by pcr technique. the obtained data were analyzed using spss ν21 software and logistic regression test.results: in this study, 15 different genotypes and 7 alleles were identified in the vntr polymorphism of nlrp3 gene, of which 12 replicate alleles and 12.12 genotypes were the most frequence. also, allele with 14 repeat were identified for the first time. analysis of the results showed that genotype 12.12 increase the risk of gastric cancer compared with other genotypes (p value = 0.01, ci = 0.14 - 0.76, or = 0.32).conclusion: it seems that genotype 12.12 in the intron 4 of nlrp3 gene increased the risk of gastric cancer.
