چکیده
|
زمینه و هدف: پپتیدهای نفوذکننده در سلول (cpps) میتوانند از غشای سلولی عبور کنند و با قابلیت حمل مولکولهای مختلف به درون سیتوپلاسم سلولها در زمینه رهایش دارویی، ژن درمانی و درمان سرطان مورد استفاده قرار بگیرند. نانوذرات طلای کروی (gnps) بدلیل سنتز آسان، حلالیت بالا، پراکنش نوری فوق العاده و عدم سمیت ابزار مناسبی برای تصویربرداری مولکولی میباشند. در این مطالعه از یکی از پپتیدهای مشتق از پپتید ماروکلسین (mca) سم عقرب (mcauf19-c) برای اتصال به نانوذرات طلا استفاده شد تا قابلیت نفوذ انتخابی داروی ضدسرطان متصل به سامانه پپتیدنانوذره، به ردههای سلولی سرطانی و میزان مرگ و میر سلولی حاصل از آن مورد ارزیابی قرار گیرد.روش ها: سلولهای hela و mda-mb-231 با غلظت 500 نانومولار داروی ضدسرطان دکسوروبیسین (dox) متصل به gnp (dox-gnp) و mcauf19-c-gnp (dox-mcauf19-c-gnp) تیمار شدند و میزان مرگ میر سلولی آنها با استفاده از روش mtt اندازه گیری گردید. نفوذ کیفی نانوذرات طلا به درون سلولها با استفاده از میکروسکپ زمینه تاریک و نفوذ کمی آنها با کمک طیف سنجی جذب اتمی (aas) مورد بررسی قرار گرفت.نتایج: حدود 87% از سلولهای hela بعد از تیمار با dox-gnp و doxmcauf19-c-gnp زنده باقی ماندند؛ درحالیکه درصد بقای سلولهای mda-mb-231 پس از تیمار با dox-mcauf19-c-gnp، %30 کاهش یافت. بعلاوه مطالعه با میکروسکپ زمینه تاریک نشان داد که نفوذ نانوذرات dox-mcauf19-c-gnp در سلولهای mda-mb-231 بیشتر از سلولهای hela می باشد. دادههای حاصل از طیف سنجی جذب اتمی نیز نشاندهنده 33% برداشت نانوذرات dox-mcauf19-c-gnp در سلولهای mda-mb-231 بود.نتیجهگیری: نتایج بدست آمده نشان داد سامانه پپتیدنانوذره میتواند ابزار مناسبی برای طراحی یک سیستم رهایش دارو با قابلیت نفوذ انتخابی باشد.

Effect of Peptide Derived from Scorpion Toxin on Enhanced Permeability of Doxorubicin Conjugated Gold Nanoparticles in HeLa and MDA-MB-231 Cells

Abstract
|
Background: Cell penetrating peptides (CPPs) can enter a cell through the cell membrane, and used in the fields of drug delivery, gene therapy, and cancer therapy by their property transporting various molecules into cytoplasm. Gold nanospheres (GNSs) are a useful tool for molecular imaging, because they are not cytotoxic and have high solubility, excellent light scattering property and ease of synthesis. We constructed a drug delivery system by developing gold nanospheres conjugated to MCaUF19C, a CPP derived from maurocalcine (MCa) scorpion toxin. We examined the applicability of this cellselective anticancer drug delivery system by evaluating its cellpenetrating and cell death activities.Methods: Cell viability of HeLa and MDAMB231 cells, against 500 nanomolar concentration of doxorubicin (DOX)conjugated gold nanoparticles (DOXGNPs, and DOXMCaUF1 -9CGNP) were evaluated using MTT assay. Cell penetrability of MCaUF19C peptide conjugated to gold nanoparticles investigated using of dark field imaging and atomic absorption spectroscopy (AAS).Results: HeLa cell viability was about 87% when treated with DOXGNP and DOXMCaUF19CGNP, whereas DOXMCaUF1 -9CGNP induced high cytotoxicity in MDAMB231 cells (30%). Dark field imaging demonstrated higher affinity of MCaUF1 -9CGNP for the MDAMB231 cell compared with HeLa cells. Moreover, atomic absorption spectrometry data analysis showed that 33% of the total amounts of the applied MCaUF1 -9CGNP were internalized in MDAMB231 cells. Conclusion: These results demonstrated that peptide conjugated GNP would be a useful tool for the development of a cellselective drug delivery system.
