چکیده    ﻫﺪف از ﭘﮋوﻫﺶ ﺣﺎﺿﺮ ﺗﻌﯿﯿﻦ اﺛﺮ ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﻫﻮازی ﺑﺎ و ﺑﺪون bfr ﺑﺮ ﭘﺮوﺗﺌﯿﻦﻫﺎی داﯾﻨﺎﻣﯿﮏ ﻣﯿﺘﻮﮐﻨﺪری mfn2 و drp1 و ﭘﺮوﺗﺌﯿﻦﻫﺎی ﻣﯿﺘﻮﻓﺎژی parkin و bcl2 ﻋﻀﻠﻪ اﺳﮑﻠﺘﯽ اﻧﺴﺎن ﻣﯽﺑﺎﺷﺪ. روش ﮐﺎر: در ﻣﻄﺎﻟﻌﻪ، 5 ﻣﺮد ﺟﻮان ﺳﻦ: 33/40±2/30 ﺳﺎل، وزن: 9/64±10/49ﮐﯿﻠﻮﮔﺮم، ﺷﺎﺧﺺ ﺗﻮده ﺑﺪﻧﯽ: 26/2±24/27ﮐﯿﻠﻮﮔﺮم ﺑﺮ ﻣﺘﺮ ﻣﺮﺑﻊ دو ﻣﺪاﺧﻠﻪ ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﻫﻮازی ﺑﺎ bfr (ex+bfr و ﺑﺪون bfr (ex را در دو روز ﻣﺠﺰا ﺑﻪ ﺻﻮرت ﺳﺮی اﺟﺮا ﮐﺮدﻧﺪ. ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﻫﻮازی ﺷﺎﻣﻞ 5 وﻫﻠﻪ ﻓﻌﺎﻟﯿﺖ 2 دﻗﯿﻘﻪای ﺑﺎ ﺳﺮﻋﺖ 50 ﻣﺘﺮ ﺑﺮ دﻗﯿﻘﻪ و 1 دﻗﯿﻘﻪ اﺳﺘﺮاﺣﺖ ﺑﯿﻦ ﻫﺮ وﻫﻠﻪ ﺑﻮد ﮐﻪ ﺑﺎ و ﺑﺪون bfrاﺟﺮا ﺷﺪ. ﺑﻼﻓﺎﺻﻠﻪ ﭘﺲ از ﻓﻌﺎﻟﯿﺖ آزﻣﻮدﻧﯽﻫﺎ در وﺿﻌﯿﺖ اﺳﺘﺮاﺣﺖ در ﺗﺨﺖ ﭘﺰﺷﮑﯽ ﻗﺮار ﮔﺮﻓﺘﻪ و دوﻣﯿﻦ ﻧﻤﻮﻧﻪ ﺧﻮﻧﯽ، و ﺑﻌﺪ از 60 دﻗﯿﻘﻪ و 120 دﻗﯿﻘﻪ ﭘﺲ از ﻓﻌﺎﻟﯿﺖ دوﺑﺎر دﯾﮕﺮ ﻧﻤﻮﻧﻪ ﺧﻮﻧﯽ از آزﻣﻮدﻧﯽﻫﺎ ﮔﺮﻓﺘﻪ ﺷﺪ. ﺳﺮاﻧﺠﺎم 3 ﺳﺎﻋﺖ ﭘﺲ از ﻓﻌﺎﻟﯿﺖ ﺑﺎﯾﻮﭘﺴﯽ دوم ﺑﺎ 3 ﺳﺎﻧﺘﯽ ﻣﺘﺮ ﻓﺎﺻﻠﻪ از ﻣﮑﺎن ﺑﺎﯾﻮﭘﺴﯽ اول از آزﻣﻮدﻧﯽﻫﺎ ﺑﻪ ﻋﻤﻞ آﻣﺪ. در ﮔﺮوه ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﺑﺎ ﻣﺤﺪودﯾﺖ ﺟﺮﯾﺎن ﺧﻮن pressure cuff در ﺑﺎﻻﺗﺮﯾﻦ ﻧﻘﻄﻪ ﭘﺎی ﻫﺮ آزﻣﻮدﻧﯽ در ﻧﺎﺣﯿﻪ ران ﺑﺴﺘﻪ ﺷﺪ. ﺑﺮای ﮔﺮم ﮐﺮدن در ﮔﺮوه ﻓﻌﺎﻟﯿﺖ ورزﺷﯽ ﺑﻪ ﻫﻤﺮاه ﻣﺤﺪودﯾﺖ ﺟﺮﯾﺎن ﺧﻮن، آزﻣﻮدﻧﯽﻫﺎ ﺑﺎ ﺑﺴﺘﻦ ﺷﺮﯾﺎن ﺑﻨﺪ ﺑﺮروی ﺻﻨﺪﻟﯽ ﻧﺸﺴﺘﻨﺪ. ﻣﺤﺪودﯾﺖ ﺟﺮﯾﺎن ﺧﻮن ﺑﻪ ﻣﺪت 30 ﺛﺎﻧﯿﻪ ﺣﻔﻆ ﺷﺪ و ﺳﭙﺲ ﺑﻪ ﻣﺪت 10 ﺛﺎﻧﯿﻪ ﺑﺮداﺷﺘﻪ ﺷﺪ. اﯾﻦ ﺣﺎﻟﺖ ﺗﺎ زﻣﺎﻧﯽ ﮐﻪ ﻓﺸﺎر ﺷﺮﯾﺎن ﺑﻨﺪ از ﻓﺸﺎر اوﻟﯿﻪ 120 ﻣﯿﻠﯽﻣﺘﺮ ﺟﯿﻮه ﺑﻪ 160 ﻣﯿﻠﯽﻣﺘﺮ ﺟﯿﻮه ﺑﺮﺳﺪ، ﺗﮑﺮار ﺷﺪ. ﺑﺮای اﻧﺪازهﮔﯿﺮی ﻣﻘﺎدﯾﺮ ﭘﺮوﺗﺌﯿﻨﯽ mfn2 و bcl2 ،drp1 و parkin ﻋﻀﻠﻪ اﺳﮑﻠﺘﯽ از روش وﺳﺘﺮن ﺑﻼت اﺳﺘﻔﺎده ﺷﺪ. ﺗﺤﻠﯿﻞ دادهﻫﺎ ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮنﻫﺎی آﻧﺎﻟﯿﺰ وارﯾﺎﻧﺲ ﺑﺎ اﻧﺪازهﮔﯿﺮیﻫﺎی ﺗﮑﺮاری و t ﻫﻤﺒﺴﺘﻪ در ﺳﻄﺢ ﻣﻌﻨﯽ داری 0/05 اﻧﺠﺎم ﺷﺪ. ﻫﻤﭽﻨﯿﻦ، ﺑﺎﯾﭙﻮﺳﯽ و ﻧﻤﻮﻧﻪ ﮔﯿﺮی از ﺑﺨﺶ ﺟﺎﻧﺒﯽ ﻋﻀﻠﻪ ﭘﻬﻦ ﺟﺎﻧﺒﯽ ﮔﺮﻓﺘﻪ ﺷﺪ. ﯾﺎﻓﺘﻪ ﻫﺎ: ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﮐﻪ ﻫﺮ دو ex + bf و ex ﺑﻪ ﻃﻮر ﻗﺎﺑﻞ ﺗﻮﺟﻬﯽ ﺑﺎﻋﺚ اﻓﺰاﯾﺶ mfn2 و ﭘﺎرﮐﯿﻦ و ﻫﻤﭽﻨﯿﻦ ﮐﺎﻫﺶ drp1 را ﻧﺴﺒﺖ ﺑﻪ ﻗﺒﻞ از آزﻣﻮن ﺷﺪ 0/05

Abstract    aim: the purpose of this study was to investigate the effect of aerobic exercise with and without bfr on mitochondrial dynamic (mfn2 &drp1 and mitophagy proteins (parkin &bcl2 of human skeletal muscle.method: . in a cross sectional study, five young men (age: 33.4 ± 2.30 year, weight: 79.64 ± 10.49 kg, bmi: 26.24 ± 2.27kg/cm2 performed two aerobic exercise intervention with bfr (ex+bfr and without bfr (ex in two separate days. the aerobic exercise included 5 bouts of 2 min activities, interspersed by 1 min active rest which performed with and without bfr. immediately after the subjects' activity, they were rested in the medical bed and the second blood sample was taken, followed by 60 minutes and 120 minutes after the second activity again. finally, 3 hours after the second biopsy, 3 cm away from the first biopsy site, the subjects were evaluated. the biopsy was taken from the lateral part of the vastus lateralis muscle. western blot method was used to measure the protein levels of mfn2, drp1, bcl2 and parkin of skeletal muscle. data analysis was performed using t test at a significant level of 0.05. results: the results of the data analyze showed that both ex+bf and ex significantly increased mfn2 and parkin and also significantly decreased drp1 compared to the pre test (p<0.05 . however, ex+bf resulted in a significant increase protein levels of bcl2 compared to the pre test (p<0.05 . also, ex+bf has a significant effect on mfn2, bcl2 and parkin elevation and drp1 decrease in compared to ex (p<0.05 .conclusion: based on the findings of this study, it seems that aerobic exercise with and without bfr is a strong stimulant for the improvement of mitochondrial dynamics and mitophagy of skeletal muscle.
