ﻫﺪف ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ، ﺑﺮرﺳﯽ اﺛﺮ ﺗﻤﺮﯾﻦ ﻫﻮازی ﺑﺮ ﺑﯿﺎن lncrna ﻫﺎی ﻣﺮﺗﺒﻂ ﺑﺎ ﮔﻠﻮﮐﺰ 6 ﻓﺴﻔﺎﺗﺎز g6p و ﮔﻠﻮﮐﻮﮐﯿﻨﺎز gck در ﻣﻮشﻫﺎی دﯾﺎﺑﺘﯽ ﺑﻮد. روش ﮐﺎر: ﻫﺠﺪه ﺳﺮ ﻣﻮش 6/c57bl ﺑﺎ وزن 12 18 ﮔﺮم و ﺳﻦ 6 4 ﻫﻔﺘﻪ ﺑﻪ دو ﮔﺮوه ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ: 1 ﻣﻮشﻫﺎی ﺗﻐﺬﯾﻪ ﺷﺪه ﺑﺎ رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب n=12 hfd و 2 ﻣﻮشﻫﺎی ﺗﻐﺬﯾﻪ ﺷﺪه ﺑﺎ رژﯾﻢ ﻏﺬاﯾﯽ اﺳﺘﺎﻧﺪارد n=6 . ﺣﯿﻮاﻧﺎت ﺑﻪ ﻣﺪت 16 ﻫﻔﺘﻪ ﺗﺤﺖ ﺗﯿﻤﺎر رژﯾﻢ ﻏﺬاﯾﯽ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﭘﺲ از ﺗﺎﯾﯿﺪ اﻟﻘﺎی دﯾﺎﺑﺖ ﺗﻮﺳﻂ ﺗﺴﺖﻫﺎی ﺗﺤﻤﻞ ﮔﻠﻮﮐﺰ و ﺗﺤﻤﻞ اﻧﺴﻮﻟﯿﻦ، ﮔﺮوه ﺗﻐﺬﯾﻪ ﺷﺪه ﺑﺎ hfd ﺑﻪ دو ﮔﺮوه ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ: a رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب ﺗﻤﺮﯾﻦ، b رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب ﺑﯽ ﺗﺤﺮک. ﻣﻮشﻫﺎی ﮔﺮوه ﺗﻤﺮﯾﻨﯽ ﺑﻪ ﻣﺪت 8 ﻫﻔﺘﻪ، 5 ﺟﻠﺴﻪ در ﻫﻔﺘﻪ ﺑﺎ ﺳﺮﻋﺖ ﻣﺘﻮﺳﻂ 22 ﻣﺘﺮ ﺑﺮ دﻗﯿﻘﻪ و ﺑﻪ ﻣﺪت 50 دﻗﯿﻘﻪ، ﺗﺤﺖ ﺗﻤﺮﯾﻦ ﺑﺮ روی ﺗﺮدﻣﯿﻞ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. 24 ﺳﺎﻋﺖ ﭘﺲ از آﺧﺮﯾﻦ ﺟﻠﺴﻪ ﺗﻤﺮﯾﻨﯽ و ﭘﺲ از 12 ﺳﺎﻋﺖ ﻧﺎﺷﺘﺎﯾﯽ، ﻣﻮشﻫﺎ ﻗﺮﺑﺎﻧﯽ و ﺑﺎﻓﺖ ﮐﺒﺪ ﺟﺪاﺳﺎزی ﺷﺪ. ﺑﺮاﺳﺎس ﻧﺘﺎﯾﺞ ﺑﯿﻮاﻧﻔﻮرﻣﺎﺗﯿﮏ، lncrna meg3 ﻣﺮﺗﺒﻂ ﺑﺎ g6p و lncrna lnclgr ﻣﺮﺗﺒﻂ ﺑﺎ gck اﻧﺘﺨﺎب ﺷﺪﻧﺪ. از آزﻣﻮن anova اﺳﺘﻔﺎده ﺷﺪ. ﯾﺎﻓﺘﻪ ﻫﺎ: ﺳﻄﻮح g6p و meg3 در ﻣﻮشﻫﺎی hfd ﺑﺎﻻ و در ﻣﻮشﻫﺎی ﺗﻤﺮﯾﻦ ﮐﺮده ﻧﺴﺒﺖ ﺑﻪ ﻣﻮش ﻫﺎی ﺗﻤﺮﯾﻦ ﻧﮑﺮده ﺑﻪ ﻃﻮر ﻣﻌﻨﺎداری ﮐﻤﺘﺮ ﺑﻮد. ﻫﻤﭽﻨﯿﻦ ﺑﯿﺎن gck و lnclgr در ﻣﻮشﻫﺎی دﯾﺎﺑﺘﯽ ﺗﻤﺮﯾﻦ ﻧﮑﺮده ﮐﻤﺘﺮ ﺑﻮد، اﻣﺎ ﺑﻪ دﻧﺒﺎل ﺗﻤﺮﯾﻦ ﺑﺪﻧﯽ اﻓﺰاﯾﺶ ﯾﺎﻓﺖ. ﻧﺘﯿﺠﻪ ﮔﯿﺮی: اﺣﺘﻤﺎﻻ ﺗﻤﺮﯾﻦ ﻫﻮازی ﺑﺎ ﮐﺎﻫﺶ ﺑﯿﺎن meg3، ﺑﯿﺎن g6p در ﮐﺒﺪ را ﺗﻨﻈﯿﻢ ﻣﯽﮐﻨﺪ و در ﻧﺘﯿﺠﻪ ﮔﻠﻮﮐﻮﻧﺌﻮژﻧﺰ و ﺗﻮﻟﯿﺪ ﮔﻠﻮﮐﺰ ﮐﺒﺪی ﮐﺎﻫﺶ ﭘﯿﺪا ﻣﯽﮐﻨﺪ و ﻣﻘﺎوﻣﺖ ﺑﻪ اﻧﺴﻮﻟﯿﻦ در ﻣﻮشﻫﺎی دﯾﺎﺑﺘﯽ ﺑﻬﺒﻮد ﻣﯽﯾﺎﺑﺪ. ﻫﻤﭽﻨﯿﻦ ﺑﻪ دﻧﺒﺎل ﺗﻤﺮﯾﻦ ﺑﺪﻧﯽ و اﻓﺰاﯾﺶ ﺑﯿﺎن lnclgr، ﺑﯿﺎن gck ﮐﺒﺪی ﻧﯿﺰ ﺑﺎﻻ ﻣﯽ رود. در اﯾﻦ وﺿﻌﯿﺖ ﮔﻠﻮﮐﺰ ﺧﻮن ﺑﻪ ﮔﻠﯿﮑﻮژن در ﮐﺒﺪ ذﺧﯿﺮه ﻣﯽﺷﻮد و در ﻧﺘﯿﺠﻪ ﻫﺎﯾﭙﺮﮔﻼﯾﺴﻤﯽ ﮐﺎﻫﺶ ﻣﯽﯾﺎﺑﺪ.

Abstract
|
aim: the aim of this study was to investigate the effect of aerobic exercise on the expression of lncrnas associated with glucose 6 phosphatase g6p and glucokinase gck in obese diabetic mice.method: eighteen c57bl/6 mice with weight 12 18 g and age 4 6 weeks were divided into two groups: mice fed a high fat diet hfd n=12 and mice fed a standard diet n=6 . the animals for 16 weeks were treated with diet. after confirmation of induction of diabetes by glucose tolerance and insulin tolerance tests, the hfd fed group was divided into two groups: a high fat diet exercise, b high fat diet sedentary. the exercised mice were trained on a treadmill for 8 weeks, 5 sessions per week at an average speed of 22 m/min for 50 minutes. twenty four hours after the last training session and after 12 hours of fasting, the mice were sacrificed and the liver tissue was isolated. based on bioinformatics results, meg3 lncrna associated with g6p and lncrna lnclgr associated with gck were selected. anova test was used.results: the levels of g6p and meg3 were significantly higher in the hfd fed mice and significantly lower in trained mice compared to untrained mice. the expression of gck and lnclgr were also lower in untrained diabetic rats, but increased after exercise.conclusion: aerobic exercise may regulate g6p expression in the liver by reducing meg3 expression, resulting in decreased gluconeogenesis and hepatic glucose production and improved insulin resistance in the diabetic mice. the expression of hepatic gck also increases following exercise and increased lnclgr expression. in this condition, blood glucose is stored in glycogen in the liver, resulting in reduced hyperglycemia.
