چکیده
|
ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل (crc) ﯾﮑﯽ از ﻣﻬﻤﺘﺮﯾﻦ ﺳﺮﻃﺎنﻫﺎ و دوﻣﯿﻦ ﻋﻠﺖ ا ﺻﻠﯽ ﻣﺮگوﻣﯿﺮ ﻧﺎ ﺷﯽ از ﺳﺮﻃﺎن در اﯾﺮان ا ﺳﺖ. اﯾﻦ ﺳﺮﻃﺎن از ﻃﺮﯾﻖ ﺗﻐﯿﯿﺮات ژﻧﺘﯿﮏ و اﭘﯽژﻧﺘﯿﮏ ﮔ ﺴﺘﺮش ﻣﯽﯾﺎﺑﺪ. ﺷﻨﺎﺧﺖ ﻣ ﺴﯿﺮﻫﺎی ﻣﻮﻟﮑﻮﻟﯽ در اﯾﻦ ﺗﻐﯿﯿﺮات ژﻧﺘﯿﮑﯽ و اﭘﯽ ژﻧﺘﯿﮑﯽ ﻣﯽﺗﻮاﻧﺪ ﭼ ﺸﻢ اﻧﺪاز رو ﺷﻨﯽ را در درﻣﺎن ﺳﺮﻃﺎن اﯾﺠﺎد ﮐﻨﺪ. اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺑﺮروی 40 ﻧﻤﻮﻧﻪ زواﺋﺪ اوﻟﯿﻪ ﺗﻮﻣﻮری روده )ﭘﻮﻟﯿﭗ( ، 30 ﻧﻤﻮﻧﻪ ﺗﻮﻣﻮری و40 ﺑﺎﻓﺖ ﻧﺮﻣﺎل ﻣﺠﺎور ﺗﻮﻣﻮر اﻧﺠﺎم ﺷﺪ. اﺳﺘﺨﺮاج dna rna ﺑﺎﻓﺘﯽ، ﺑﺮ ﺳﯽ ﺑﯿﺎن ژنﻫﺎی 194-mir-22 ,mir, real time pcr lncrna mincr اﻧﺠﺎم ﺷﺪ. در اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﻣﯿﺰان ﺑﯿﺎن lncrna mincr در ﻧﻤﻮﻧﻪﻫﺎی ﺗﻮﻣﻮر و ﭘﻮﻟﯿﭗ ﻧﺴﺒﺖ ﺑﻪ ﺑﺎﻓﺖ ﻧﺮﻣﺎل ﻣﺠﺎور ﺷﺎن اﻓﺰاﯾﺶ و ﻣﯿﺰان ﺑﯿﺎن 194-mir و mir-22 وکاهش یافته بود. بین میزان بیان,mir-22 ,mir-194 و mincr ضریب همبستگی منفی مشاهده شد و همچنین بین بیان این lncrna و microrna ها ارتباط معنی داری مشاهده می شود. mincr ﯾﮑﯽ از ﺟﺪﯾﺪﺗﺮﯾﻦ lncrna ﻫﺎی ﺷﻨﺎ ﺳﺎﯾﯽ ﺷﺪه ﻣﯽﺑﺎ ﺷﺪ ﮐﻪ ﺗﺎﮐﻨﻮن در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﺑﺮر ﺳﯽ ﻧ ﺸﺪه و ﻣ ﺴﯿﺮﻫﺎی ﺳﯿﮕﻨﺎﻟﯿﻨﮓ آن ﺑﻪ در ﺳﺘﯽ ﺑﺮر ﺳﯽ ﻧ ﺸﺪه ا ﺳﺖ. واﺑ ﺴﺘﻪ ﺑﻮدن ﺑﯿﺎن mincr ﺑﺎ 22-mir-194 mir ﺑﻪﻋﻨﻮان ﯾﮑﯽ از ﻣﺴﯿﺮﻫﺎی ﺳﯿﮕﻨﺎﻟﯿﻨﮓ ﻣﯽﺑﺎﺷﺪ ﮐﻪ اوﻟﯿﻦ ﺑﺎر در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﺑﺮرﺳﯽ ﯿﺎن اﯾﻦ microrna lncrna ﮐﻪ ﻫﻤﺒﺴﺘﮕﯽ ﻣﻌﻨﺎ داری ﻧﯿﺰ ﻣﯽﺑﺎﺷﺪ در ﻧﻤﻮﻧﻪﻫﺎی ﭘﻮﻟﯿﭗ و ﺗﻮﻣﻮر ﺑﺮای اوﻟﯿﻦ ﺑﺎر در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﺑﺮرﺳﯽ ﺷﺪه اﺳﺖ. ﺗﮑﻤﯿﻞ ﻣﺴﯿﺮﻫﺎی ﺳﯿﮕﻨﺎﻟﯿﮏ ﻣﯽﺗﻮاﻧﺪ اﻣﯿﺪاواری در ﺗ ﺸﺨﯿﺺ و درﻣﺎن را در اﻧﻮاع ﻣﺨﺘﻠﻒ ﺳﺮﻃﺎنﻫﺎ در ﭘﯽ دا ﺷﺘﻪ ﺑﺎ ﺷﺪ. mincr ﯾﮑﯽ از ﺟﺪﯾﺪﺗﺮﯾﻦ lncrnaﻫﺎی ﺷﻨﺎﺳﺎﯾﯽ ﺷﺪه ﻣﯽﺑﺎﺷﺪ ﮐﻪ ﺗﺎﮐﻨﻮن در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﺑﺮرﺳﯽ ﻧﺸﺪه و ﻣﺴﯿﺮﻫﺎی ﺳﯿﮕﻨﺎﻟﯿﻨﮓ آن ﺑﻪ درﺳﺘﯽ ﺑﺮرﺳﯽ ﻧﺸﺪه اﺳﺖ.

Abstract
|
colorectal cancer (crc) is the third most common cancer after lung and breast cancer and the fourth leading cause of oncological death in the world. great efforts have been made to develop effective methods for crc diagnosis and treatment. long non-coding rnas (lncrnas) have been demonstrated to have important roles in gene regulation and numerous lncrnas have been discovered. they have been found to be important regulators of gene expression in development, physiology and when dysfunctional in the presence of disease.this study was conducted on 40 samples of polyps, 30 tumor samples and 40 normal tissue adjacent to the tumor. rna and tissue dna extraction, mir-22, mir-194 lncrna mincr gene expression analysis was performed using real time pcr method. the expression level of lncrna mincr in tumor and polyp samples was increased compared to the adjacent normal tissue, and the expression level of mir-194, mir-22, and decreased. a negative correlation coefficient was observed between the expression levels of mir-22, mir-194 and mincr, and a significant correlation was also observed between the expression of these lncrnas and micrornas.mincr is one of the most recently identified lncrnas that have not been investigated in colon cancer and its signaling pathways have not been properly investigated. the dependence of mincr expression with mir-22 and mir-194 is one of the signaling pathways that has been investigated for the first time in colon cancer. the dependence of the expression of this lncrna and microrna, which is also a significant correlation, has been investigated in polyp and tumor samples for the first time in colon cancer. completing signaling pathways can lead to hope in the diagnosis and treatment of various types of cancer. mincr is one of the most recently identified lncrnas that have not been investigated in colon cancer and its signaling pathways have not been properly investigated.
