چکیده
|
علفهای هرز از جمله خردل وحشی یک مسئله جدی در تولید محصولات زراعی هستند. مقاومت به علفکشها در علفهای هرز و خردل وحشی به یک تهدید متداول در کشاورزی تبدیل شده است و امنیت غذای جهانی را به خطر انداخته است. در این آزمایش ژن als علف هرز خردل وحشی مشکوک به مقاومت ناشی از غربال مزرعهای و روشهای مبتنی بر pcr از مزارع گندم استان کرمانشاه مورد بررسی قرار گرفت. سپس ژن als شناسایی، جداسازی و همسانهسازی شد. برای این منظور با توجه به اطلاعات موجود در پایگاه ncbi، پرایمرهای مناسب طراحی و با rtpcr توالی ژنی مذکور تکثیر و به منظور توالییابی در وکتور ptz57r/t درون باکتری e. coli سویهی dh5 alpha; همسانهسازی شد. براساس نتایج مولکولی مبتنی بر pcr، تعداد شش بیوتیپ، جهش در جایگاه هدف را نشان دادند. نتایج توالییابی براساس روش مبتنی بر pcr، جهشهای نوکلئوتیدی asp376gly (gac به gag)، pro197ser (cct به tct)، trp574leu (tgg به ttg) و ala122thr (gca به aca) در شش بیوتیپ را تایید نمود. این جایگاههای جهش توسط روش توالییابی تایید گردید. براساس نتایج این تحقیق مقاومت به علفکش در گیاه خردل وحشی میتواند بهدلیل مقاومت در جایگاه هدف بهدلیل جهشهای نوکلئوتیدی مذکور در جایگاه فعال آنزیم، محل اتصال علفکشهای بازدارنده als و خانواده آن باشد، یا بهدلیل مقاومت جایگاه هدف که با تغییرات کنفورماسیونی میتواند باعث تغییراتی در سطح جایگاه واکنش پروتئین با علفکش و در نتیجه تغییر در میزان فعالیت و اتصال آنزیم علفکش شود. این بدان معنی است که سطح مولکولی آنزیم نوع موتانت، کمتر در معرض تماس با علفکش قرار میگیرند و علفکش نمیتواند آنزیم را از فعالیت باز دارد و باعث ایجاد مقاومت در برابر علفکش میشود.

Genetic and molecular study of resistance in weed wild mustard (Sinapis arvensis) to acetolactate synthase inhibitor herbicide

Abstract
|
Weeds such as wild mustard are a serious problem in crop production. Herbicide resistance in weeds and wild mustard has become a common threat in agriculture and endangers global food security. In this experiment, the ALS gene of wild weed suspected of weeding due to field screening and PCRbased methods from Kermanshah province #39;s wheat fields was investigated. The ALS gene was then identified, isolated and cloned. For this purpose, primers were designed according to the information available in the NCBI database and ALS gene was amplified using RTPCR. The gene ALS cloned in pTZ57R / T vector within E. coli bacteria, DH5 alpha; strain and sequenced. Based on PCRbased molecular results, six biotypes showed mutations in the target site. PCRbased sequencing results confirm the nucleotide mutations of Asp376Gly (GAC to GGC), Pro197Ser (CCT to TCT), Trp574Leu (TGG to TTG), and Ala122Thr (GCA to ACA) in six biotypes. These mutation sites were confirmed by sequencing. According to the study, herbicide resistance in wild mustard could be related to resistance in the target site due to the mentioned nucleotide mutations in the active site of the enzyme, the binding site of ALS inhibitors and its family, or due to the resistance of the target site that can cause conformational changes. Changes in the level of the protein reaction site with herbicides and as a result changes in activity and enzymeherbicide binding. This means that the molecular levels of the mutant enzyme are less likely to come into contact with herbicides, and the herbicide cannot stop the enzyme from working and cause herbicide resistance.
