بررسی سطح بیان ژن ve-cadherin در سرم زنان ایرانی مبتلابه سرطان سینه

زمینه و هدف: سرطان سینه یکی از شایع‌ترین سرطان‌ها و دومین عامل مرگ ناشی از سرطان زنان در جهان هم در کشورهای توسعه‌یافته و هم در کشورهای درحال‌توسعه است. مولکول ve-cadherin، مولکول اندوتلیالی مخصوص اتصال است که در محل‌های اتصال سلول‌های اندوتلیال قرار دارد و ارتباط آن با سرطان‌های مختلف اثبات‌شده است. هدف از مطالعه حاضر، بررسی میزان بیان ژن ve-cadherin در سرم بیماران ایرانی مبتلابه سرطان سینه به‌منظور شناسایی بیومارکری در پیش‌آگاهی آن بود.مواد و روش‌ها: بعد از جداسازی سرم از نمونه‌های خون بیماران و کنترل، استخراج rna و سنتز cdna انجام شد. بیان ژن با تکنیک realtime pcr اندازه‌گیری شد.نتایج: نتایج نشان داد که بیان ژن ve-cadherin در نمونه‌های بیمار در مقایسه با کنترل در سطح معنی‌داری افزایش‌یافته است؛ اما ارتباطی بین میزان بیان و درجه تومور به دست نیامد.نتیجه‌گیری: با توجه به نتایج به‌دست‌آمده در این مطالعه، بررسی بیان این فاکتور می‌تواند به‌عنوان مارکری جهت بررسی و فالوآپ بیماران مبتلابه سرطان سینه باشد. همچنین با توجه به نقش این فاکتور در رگ زایی و متاستاز تومورها، می‌توان از آن به‌عنوان هدفی برای مداخله‌ی درمانی استفاده کرد.

The Evaluation of VE-Cadherin Gene Expression in Serum of Iranian Women With Breast Cancer

Background Objective: Breast cancer is one of the most common cancers and the second leading cause of cancer death in women worldwide, in both developed and developing countries. Vascular endothelial (VE)cadherin is endothelial specific adhesion molecule located at junctions between endothelial cells and its association with various cancers has been proven. The aim of this study was to evaluate the VEcadherin gene expression in serum of Iranian women with breast cancer in order to identify a biomarker for its prediction.Materials Methods: After serum isolation from blood samples of the patients and controls, extraction of RNA and synthesis of cDNA were done. Gene expression was measured using realtime PCR technique.Results: Our results showed that the expression of VEcadherin was increased significantly in patients compared to control samples. However, there was no relationship between its expression and grade of the tumors.Conclusion: Based on the results of this study, expression of this factor can be used as a marker for study and followup of breast cancer patients. Furthermore, considering the role of this factor in angiogenesis and metastasis, it can be used as a target for intervention therapy.
