   انتقال و بیان ژن اکسندین- 4 متصل به زیر واحد b سم وبا ( ctb ) در گیاه کاهو

چکیده    اکسندین-4 (ex4) یک پروتئین شبیه به glp-1 است با این تفاوت که دارای نیمه عمر بیشتر و در نتیجه تاثیر بیشتر در القا ترشح انسولین است. اتصال ex4 به زیر واحد غیر سمی وبا یا ctb موجب اقزایش کارایی آن از طریق جذب در سیستم گوارشی می شود. در این تحقیق، ناقلpbi121 دارای قطعه ctb-ex4 توسط باکتری agrobacterium tumefaciens به هسته گیاه کاهو معرفی شد. 20.25% از ریزنمونه‎‌های تلقیح شده با موفقیت تراریخت و باززا شدند و پس از تایید در آزمون pcr، بذرهای تراریخت نسل اول (t0) از گیاهان حاصل از این ریز نمونه ها تهیه گردید. برای ایجاد گیاهان نسل دوم (t1) در مجموع تعداد 24 عدد بذر t0 کشت گردید که 42% از بذرهای کشت شده به صورت کامل رشد کردند و به بذر نشستند. با استفاده از آغازگرهای اختصاصی در واکنش pcr، تراریختی و حضور ژن ctb-ex4 در گیاهان t1 مورد بررسی شد. بیان نیمه کمی و کمی ژن ctb-ex4 در سطح mrna با استفاده از آزمون های rt-pcr و real time pcr و در سطح پروتئین با استفاده از آنتی‌بادی‌های اختصاصی و آزمون‌های elisa و western blot مورد بررسی قرار گفت. در نهایت تعداد نسخه های انتقال یافته به گیاهان t1 با استفاده از دستگاه real time pcr با استفاده از روش مطلق و منحنی استاندارد و همچنین روش نسبی، نسبت به ژن کنترل داخلی actin برآورد شد. نتایج حاکی از تراریختی 60% لاین‌های نسل دوم (t1) و بیان ژن ctb-ex4 در سطح mrna و پروتئین بود. نتایج نشان داد که مقدار بیان در لاین های مورد بررسی متفاوت بود. بیشترین مقدار بیان در سطوح پروتئین و mrna در لاین 105 مشاهده شد. بیشترین تعداد نسخه منتقل شده در لاین 101 و کمرین تعداد نسخه منتقل شده به میزان 1 نسخه در لاین های 104 و 105 مشاهده شد. نتایج حاصل نشان داد که پروسه انتقال ژن ctb-ex4 به گیاه کاهو موفقیت‌آمیز بود وگیاه کاهو قادر به بیان ژن مورد نظر در سطوح rna و پروتئین بود. با در نظر گرفتن پروسه‌های بعدی از جمله تخلیص و ارزیابی فعالیت بیولوژیکی پروتئین تولید شده، از گیاهان می‌توان به عنوان یک بیوراکتور مناسب جهت تولید پروتئین‌های نوترکیب بهره برد.

   transfer and expression of exendin-4 fused to cholera toxin b subunit (ctb) in lettuce

Abstract    exendin-4 (ex4) is a protein similar to glucan-like peptide (glp-1) and has a longer half-life and greater effect in inducing insulin secretion. the fusion of ex4 to the non-toxic cholera subunit (ctb) increases its efficiency through absorption in the digestive system. in this study, ctb-ex4 gene was introduced into lettuce by agrobacterium tumefaciens using vector pbi121. the results showed that 20.25% of the inoculated explants were successfully transformed and regenerated (t0 generation). to create t2 plants, seeds of t1 were cultivated, and 42% of the cultivated seeds germinated. t1 plants were analyzed using pcr test. the mrna expression of the ctb-ex4 gene was examined through rt-pcr and real-time pcr, while the protein expression was assessed using elisa and western blot tests. the number of transferred copies to t1 plants was determined using real-time pcr. this estimation was performed using both the absolute method and the standard curve, as well as the relative method, with acitn gene as a reference gene. the results showed that 60% of the t1 plants were transgenic and the ctb-ex4 gene was expressed at the mrna and protein levels. the results showed that the process of transferring the ctb-ex4 gene to the lettuce plant was successful and the lettuce plant was able to express the desired gene at the rna and protein levels. considering the subsequent processes, including the purification and evaluation of the biological activity of the produced protein, plants can be used as a suitable bioreactor for the production of recombinant proteins.
