   بررسی وابستگی ژنوتیپی باززایی درون‌شیشه‌ای گیاه بنفشه آفریقایی

چکیده    بنفشه آفریقایی یکی از گیاهان زینتی محبوب با تنوع بسیار بالا می باشد، که به دلیل عدم وجود سیستم تولیدمثل جنسی مناسب، تکثیر و تولید آن از روش های غیرجنسی انجام می شود. با وجود تنوع زیاد ژنوتیپی در این گیاه، مطالعه ای روی وابستگی باززایی آن در شرایط درون شیشه ای به ژنوتیپ گزارش نشده است. در این پژوهش، پانزده اکوتیپ مختلف با تفاوت های برجسته ظاهری از نظر پتانسیل باززایی درون شیشه ای روی محیط کشت بهینه شده، کشت شدند. نتایج آزمایش ها نشان داد که همه ژنوتیپ های بررسی شده قابلیت باززایی روی این محیط را دارند که به غیر از دو اکوتیپ، بقیه در سه گروه آماری نزدیک به هم قرار گرفتند. این نتایج حفظ شدن عوامل اصلی دخیل در پتانسیل باززایی را در ژنوتیپ های مختلف بنفشه آفریقایی در طول زمان اثبات می کند. این حفاظت می تواند به نیاز این گیاه به روش های تکثیر جایگزین به دلیل عدم وجود سیستم تولید مثل جنسی مناسب مرتبط باشد. با این حال، تعداد گیاهچه های باززا شده از هر ریزنمونه در اکوتیپ های مختلف به شدت متفاوت بود، که می تواند به اثر متقابل سایر شرایط محیطی علاوه بر هورمونها در فعال سازی عوامل ژنتیکی و اپی ژنتیکی لازم برای هدایت سرنوشت سلول ها به سمت باززایی گیاه از طریق یکی از مکانیسم های جنین زایی سوماتیکی یا اندام زایی مربوط باشد. به طور کلی می توان گفت در گیاه بنفشه آفریقایی مکانیسم های باززایی گیاه در تمام ژنوتیپ ها حفظ شده است، با این حال، فعال سازی آنها در ژنوتیپ های مختلف برای افزایش تعداد گیاهچه ها در واحد سطح ریزنمونه به بهینه سازی عوامل محیطی وابسته است.

   investigating the genotype dependency of in vitro regeneration of african violet

Abstract    african violet is one of the popular ornamental plants with high diversity, that is propagated through asexual methods due to the lack of a proper sexual reproduction method. despite the great genotypic diversity in this plant, no study has been reported on the genotype dependency of its in vitro regeneration. in this research, to evaluate in vitro regeneration potential, fifteen different african violet ecotypes with prominent phenotypic differences were cultured on the optimized regeneration medium. the results showed that all the studied genotypes have the ability to regenerate on this medium and, except for two ecotypes, the rest were placed in three statistical groups close to each other. these results prove the preservation of major factors involved in regeneration potential in different genotypes of african violets over time. this conservation can be related to the need of this plant for alternative reproduction methods due to the lack of an accurate sexual reproduction system. however, the number of regenerated seedlings from each explant was highly different in various ecotypes, which could be due to the interaction of other environmental conditions in addition to hormones in the activation of genetic and epigenetic factors necessary to prime cell fate towards plant regeneration through one of the mechanisms of somatic embryogenesis or organogenesis. in general, it can be concluded that in the african violet plant, regeneration mechanisms have been preserved in all genotypes, however, their activation in different genotypes to increase the number of regenerants per explant is dependent on optimizing environmental factors.
