چکیده
|
سابقه و هدف: استفاده از داروهای شیمیایی بهمنظور درمان سرطان کبد دارای عوارض جانبی متعددی میباشد. ازاینرو استفاده از دارویی که با عوارض کمتر بتواند بر زندهمانی سلولهای سرطانی تاثیرگذار باشد از اهمیت بهسزایی برخوردار است. هدف از مطالعۀ حاضر، بررسی تاثیر مهاری آلفا-آمانیتین بر سلولهای hepg2 میباشد. مواد و روش ها: در این مطالعه، کشت سلولهای hepg2 بر روی پردۀ آمنیون حاوی huvec انجام شد و پس از تیمار با آلفا-آمانیتین، زندهمانی سلولها با استفاده از تست mtt و میزان آپوپتوز با استفاده از تکنیک فلوسایتومتری بررسی شد. تعیین کمیّت مالوندیآلدهید (mda)، میزان فعالیت آنزیمی سوپراُکسید دیسموتاز (sod)، وضعیت اُکسیدانی کل (tos) و ظرفیت آنتیاُکسیدانی کل (tac) با استفاده از روش کالریمتری انجام شد. ارزیابی میزان فاکتور نکروز تومور آلفا (tnf-α) با تکنیک ایمونوسیتوشیمی (icc) انجام شد. یافتهها: نتایج تست mtt نشان داد که تیمار سلولهای hepg2-amnion و hepg2-amnion-huvec با غلظتهای 250 و 500 پیکوگرم بر میلیلیتر داروی آلفا-آمانیتین باعث کاهش معنادارِ زندهمانی سلولهای hepg2 شد. همچنین نتایج فلوسایتومتری نشان داد که بروز آپوپتوز در سلولهای hepg2-amnion-huvec و hepg2-amnion تیمارشده با آلفا-آمانیتین بهطور معناداری افزایش یافته است. نتایج آزمون بیوشیمیایی افزایش میزان mda، tos و کاهش سطح sod و tac را در هر دو گروه همکِشتیشده با آمنیون و amnion-huvec و تیمارشده با آلفا-آمانیتین را نشان داد. نتایج icc نشان داد که میزان بیان پروتئین tnf-α در هر دو گروه تیمارشده با آلفا-آمانیتین افزایش معناداری داشته است. نتیجهگیری: استفاده از غشای آمنیونی برای کشت سلولهای hepg2 میتواند سبب تاثیر ضدسرطانی قابلتوجه آلفا-آمانیتین بر این نوع سلولها شود.

Abstract
|
aim and background: the use of chemical drugs to treat liver cancer has many side effects. therefore, the use of a drug that can affect the survival of cancer cells with fewer side effects is of great importance. the aim of the present study was to investigate the inhibitory effect of alpha-amanitin on hepg2 cells.materials and methods: in this study, hepg2 cells were cultured on amniotic membrane containing huvec and after treatment with alpha-amanitin, cell viability was assessed using mtt assay and apoptosis was assessed using flow cytometry. malondialdehyde (mda), superoxide dismutase (sod) enzyme activity, total oxidant status (tos) and total antioxidant capacity (tac) were quantified using calorimetry. tumor necrosis factor alpha (tnf-α) levels were assessed using immunocytochemistry (icc).results: the results of the mtt test showed that the treatment of hepg2-amnion and hepg2-amnion-huvec cells with concentrations of 250 and 500 pg/ml of alpha-amanitin significantly reduced the viability of hepg2 cells. also, the results of flow cytometry showed that the incidence of apoptosis in hepg2-amnion-huvec and hepg2-amnion cells treated with alpha-amanitin significantly increased. the results of the biochemical test showed an increase in the levels of mda, tos and a decrease in the levels of sod and tac in both groups co-cultured with amnion and amnion-huvec and treated with alpha-amanitin. the results of the icc showed that the expression of tnf-α protein in both groups treated with alpha-amanitin significantly increased.conclusion: using amniotic membrane to culture hepg2 cells can result in a significant anticancer effect of alpha-amanitin on this type of cell.
