چکیده
|
هدف: نظر به این که تغذیه بر تمامی فعل و انفعالات بدن، به ویژه بر ژنوم و نیز بیان ژنها تاثیر گذار است و همه موجودات همواره در حال تغذیه هستند و بدون تغذیه حیات امکانپذیر نیست. لذا هدف این پژوهش بررسی اجمالی نقش تغذیه در بیان ژن، همانندسازیdna ، جلوگیری از آسیب dna و ترمیمdna بود. مواد و روشها: در این مطالعه با استفاده از کلید واژههای سرطان، ریبونوکلئاز، مهارکننده ریبونوکلئازی و اسید ریبونوکلئیک، در پایگاههای اطلاعاتی معتبر scopus، sid، irandoc، pubmed، google scholar، web of science و iranmedex جستجو انجام شد. به منظور انتخاب مستندات مورد استفاده، تمام مقالاتی که به زبان غیرانگلیسی و فارسی چاپ شده بودند، مقالات تکراری، مقالاتی که امکان دستیابی به متن کامل مقاله وجود نداشت و همچنین مقالاتی که به صورت چکیده ارائه شده بودند حذف گردیدند. نهایتا موارد منتخب به منظور تنظیم مقاله حاضر به طور کامل مطالعه و خلاصهسازی شدند. نتایج: تنظیم بیان ژن برای حیات و سلامت ضروری است و به عوامل درون زا و رژیم غذایی و سایر عوامل محیطی که در سطوح مختلف و با مکانیسمهای متعدد عمل می کنند حساس است. پیوندهای مهم و نزدیکی بین رژیم غذایی/تغذیه، وضعیت متابولیک، مسیرهای سیگنالدهی و تنظیم ژن وجود دارد. فناوریهای پساژنومی شامل هیبریداسیون ریزآرایه و توالییابی نسل بعدی امروزه امکان تجزیه و تحلیل با بازدهی و خروجی بالا، در سطح گسترده ژنوم، رونویسی و پویایی آن (حضور و کمیتrna ها، الگوهای بیانrna ) را فراهم می کند. علاوه بر این، تغییرات ناشی از رژیم غذایی یا عوامل دیگر در اشغال محل اتصال فاکتورهای نسحهبرداری، ویژگیهای کروماتین، الگوهای جهانی تباهی رونویسی و پروفایل ترجمه نیز امروزه می تواند در سطح ژنوم مورد مطالعه قرار گیرد. شواهد زیادی وجود دارد که تعداد زیادی از ریز مغذیها، از جمله ویتامینها و مواد معدنی به عنوان کوفاکتورهای آنزیمها یا به عنوان بخشی از ساختار پروتئینها (متالوآنزیمها) در ترمیمdna ، جلوگیری از آسیب اکسیداتیو به dna و همچنین نگهداری و متیلاسیون dna مورد نیاز هستند. نقش ریز مغذیها در حفظ ثبات ژنوم به طور گستردهای بررسی شده است. بسیاری از ریز مغذیهای در فرآیندهای مختلف پایداری ژنوم درگیر هستند. همچنین مکانیسمهای مختلفی وجود دارند که به وسیله آنها کمبود ریز مغذی میتواند باعث آسیب بهdna ، تسریع پیر شدن و ناپایداری کروموزومی شود. نتیجه گیری: ادغام نتایج حاصل از این فناوریهای متنوع در نهایت به درک جامع و گسترده ای از سیستم کنترل بیان ژن و روابط رژیم غذاییژنسلامت در علم تغذیه مدرن کمک میکند.

Abstract
|
Objective Since nutrition affects all the interactions of the body, especially the genome as well as the expression of genes, and all organisms are always feeding, and without feeding life is not possible. Therefore, the aim of this study was to review the role of nutrition in gene expression, DNA replication, prevention of DNA damage and DNA repair. Materials and methods In this study, the keywords such as cancer, ribonuclease, ribonuclease inhibitor and RNA were used to search in databases including Scopus, SID, IranDoc, PubMed, Google Scholar, Web of Science and IranMedex. In order to select the documents used, all articles published in nonEnglish and Persian languages, duplicate articles, articles that could not be accessed to the full text, as well as articles that were presented as abstracts were removed. Finally, the selected cases were thoroughly studied and summarized in order to prepare the current review. Results Regulation of gene expression is essential to life and health and is sensitive to endogenous and dietary and other environmental factors that exert their action at multiple levels and by multiple mechanisms. There are important and intimate links between diet/nutrition, metabolic status, signaling pathways, and gene regulation. Postgenomic technologies including microarray hybridization and massive nextgeneration sequencing (RNASeq) enable nowadays highthroughput analysis, at genomewide level, of the transcriptome and its dynamics (presence and quantity of RNAs, RNA expression patterns). Further, changes induced by dietary or other factors in TF binding site occupancy, chromatin features, global patterns of transcript decay, and translational profiling can also be studied nowadays at a genomewide level. There is overwhelming evidence that a large number of micronutrients (vitamins and minerals) are required as cofactors for enzymes or as part of the structure of proteins (metalloenzymes) involved in DNA repair, prevention of oxidative damage to DNA, as well as maintenance and methylation of DNA. The role of micronutrients in the maintenance of genome stability has been extensively reviewed. Some of micronutrients involved in various genome stability processes. There are various mechanisms by which micronutrient deficiency could cause DNA damage, accelerate senescence and chromosomal instability. Conclusion Integration of results from these varied technologies will ultimately allow a comprehensive, systemwide understanding of gene expression control and of dietgenehealth relationships in modern nutrition science.
