بررسی اثرات زولپیدم بر اووژنز ومیزان fsh,lh موش بالغ نژاد nmri

هدف: این مطالعه با هدف بررسی اثرات زولپیدم بر اووژنزو هورمون‏های جنسی fsh و lh موش ماده نژاد nmri صورت گرفت. مواد و روش‏ها: در این تحقیق از 30 موش بالغ نژاد nmri با وزن تقریبی 30±26 گرم که توسط تهیه اسمیر واژنی در مرحله استروس از سیکل جنسی بودند انتخاب شدند و به گروه‏های تجربی، شاهد و کنترل تقسیم شدند. گروه‏های تجربی در 3 دسته مختلف این دارو را به‏صورت تزریق درون صفاقی به‏میزان 5، 10، 20 میلی‏گرم بر کیلوگرم وزن موش به‏مدت 14 روز دریافت نمودند گروه سم 0.05 میلی‏لیتر آب مقطر و کنترل هیچ ماده‏ای دریافت نکرد در پایان روز چهاردهم اندازه‏گیری میزان سرم هورمون‏های fsh  وlh توسط روش الیزا انجام شد و همچنین تخمدان از بدن حیوانات خارج شد ومقاطع بافتی با رنگ‏آمیزی هماتوکسیلین ائوزین تهیه شد و تحت مطالعات میکروسکوپی و هیستولوژیکی قرار گرفت و نتایج از طریق آزمون‏های  (oneway  anova)و تست tukey)) با معنی‏داری (p<0/05) تجزیه وتحلیل آماری شد. نتایج: تزریق داروی زولپیدم به‏طور معنی‏داری (p<0/05) باعث کاهش سرمی هورمون‏های fsh  و lh در گروه‏های تجربی نسبت به سم و کنترل شد، در بررسی‏های بافتی (p<0/05) افزایش فولیکول‏های بدوی، کاهش فولیکول ثانویه، گراف، جسم زرد نسبت به گروه سم و کنترل مشاهده شد همچنین کاهش معنی‏دارای در قطر فولیکول های ثانویه، گراف، جسم زرد و افزایش معنی‏داری در قطر فولیکول‏های بدوی، فولیکول‏های آترتیک در گروه های تجربی نسبت به سم و کنترل شد. نتیجه‏گیری: تزریق زولپیدم باعث تغییرات معنی‏داری در روند اووژنز و هورمون‏های fsh  و lh می‏شود.

The Effect of Zolpidem Oogenesis and LH,FSH Serum of Adult NMRI Mouse Strain

Aim: The aim of this study was the effect of zolpidem on oogenesis and FSH, LH hormones of female adult NMRI mouse. Material and Methods: In this experimental study thirty adult female mice NMRI strain at a mean weight of 30±26 grams were divided into five groups Zolpidem solution was prepared in distilled water at 5, 10, and 20 (mg/kg of body weight) doses, and 0.5 cc injections were doneintraperitoneally every day for 14 days The control group received no injection. The sham group received distilled water and treatment groups of 1, 2, and 3 received doses of 5, 10, and 20 mg/kg The treatment groups were sacrificed one day after the last injection The concentrations of the hormones were measured by the ELISA test The ovary tissue was separated and examined as well as Hematoxylin and eosin painting and the results were evaluated via the tukeytest,(Oneway ANOVA) by SPSS program. Results: The results showed a significant decrease in the mean serum FSH, LH levels in the experimental groups compared to the sham and control (P<0.05) Also histological studies of sections showed the mean number of graffian, secondary and corpus luteum reduced and the mean number of primordial, growing, atretic and abnormal follicles increased. In addition, a significant reduction in the diameter of secondary, graffian follicles and corpus luteum and a significant increased in the diameter of primary, atretic follicles were observed in the experimental groups, compared to the sham and control groups. Conclusion: Injection of zolpidem significantly is effective on , oogenesis and concentration of LH,FSH hormones.
