Prefractionation in Proteome Profile Analysis of Anxc4 Gene Mutant in Aspergillus Fumigatus

Aspergillus fumigatus is one of the pathogenic filamentous fungi that could cause opportunistic infection, allergy and poisoning. since, genetic sequencing of some of these fungi has been completed, identifying the protein profile of these fungus cells is necessary. one of these genes is the annexin family. annexin c4 is a new member of fungal annexins. this study investigates the effect of anxc4 mutant gene in proteome profile of aspergillus fumigatus. moreover, in order to enhance the power of protein complex separation, we used an optimized prefractionation method. using reverse phase-high performance liquid chromatography eight fractions were separated. then to confirm the protein concentration, each fraction was tested by sds-page. protein profile of these fractions was analyzed using 2-de, and image master software. among the proteins identified statistically, two emerging proteins were observed. the results show that the expression of anxc4 could affect the expression of some proteins in a.fumigatus. to accurately identify these proteins, further experiments are needed including mass spectrometry analysis.

Aspergillus Fumigatus در ANXC4 جداسازی جزء به جزء در تحلیل پروفایل پروتئینی موتانت ژن

Aspergillus fumigatus یکی از قارچ های رشته ای پاتوژن است که می تواند باعث عفونت های فرصت طلب ، آلرژی و مسمومیت ها شود. از آنجا که سکانس ژنی تعدادی از این قارچ ها اخیراً کامل شده است شناسایی پروفایل پروتئینی این سلول های قارچی ضروری است. یکی از این ژن ها، خانواده ی Annexin است. Annexin C4 یکی از اعضای این خانواده در قارچ ها است. در این مطالعه اثر جهش در ژن ANXC4 در پروفایل پروتئینی قارچ A. fumigatus مورد بررسی قرار گرفته است. علاوه بر این به منظور افزایش کارایی جداسازی پروتئین ها پیش از بررسی پروفایل پروتئینی تکنیک از پیش جزء به جزء کردن مطابق یک روش بهینه انجام گرفت. در این تحقیق با استفاده از کروماتوگرافی مایع فازمعکوس با کارایی بالا، 8 فراکشن جداشده سپس هرکدام از فراکشن ها با استفاده از تکنیک SDS Page برای تایید غلظت های پروتئینی روی ژل الکتروفورز نیز جداسازی شدند. پروفایل پروتئین های این سه فراکشن با استفاده از تکنیک الکتروفورز دو بعدی مورد بررسی قرار گرفته شد و با استفاده از نر افزار IMEAG MASTER نتایج آنالیز گردید. در میان پروتئین هایی که با روش آماری شناسایی گردید دو پروتئین نوظهور مشاهده گردید. نتایج نشان دادند که بیان ژن ANXC4 بیان تعدادی از پروتئین های دیگر را در قارچ A. fumigatus تحت تاثیر قرار می دهد. برای شناسایی دقیق تر این پروتئین ها آزمایش هایی از جمله آنالیز های اسپکترومتری مورد نیاز است.
