   بررسی بیان برخی از ژن‌های مقاومت به بیماری برق‌زدگی درگیاه نخود زراعی

چکیده    بیماری برق‌زدگی که به وسیله قارچ ascochyta rabiei ایجاد می‌شود، یکی از مهم‌ترین عوامل محدودکننده کشت و تولید نخود در بیشتر مناطق دنیا و از جمله ایران است. القای مقاومت نسبت به پاتوژن‌ها از راهکارهایی است که گیاهان برای مقابله با تنش‌های زیستی به کار می‌برند. این تحقیق در سال 1397 به صورت آزمایش فاکتوریل دو عاملی (زمانژنوتیپ) در قالب طرح کامل تصادفی در سه تکرار برای هر کدام از زمان‌های انتخاب شده و گیاه شاهد در آزمایشگاه گروه اصلاح نباتات دانشکده کشاورزی دانشگاه محقق اردبیلی انجام شد. به منظور درک بهتر سیستم‌های دفاعی گیاه نخود در سطح مولکولی، دو ژنوتیپ حساس (flip 03135c) و مقاوم نخود ( (flip 0040cدر گلخانه کشت شد و میزان بیان ژن‌های درگیر در مقاومت به بیماری برق‌زدگی نخود (protein with leucine zipper، snakin2،afpca وpgip ) در این ژنوتیپ‌ها مورد مطالعه قرار گرفت. برای این منظور بعد از این که گیاهان به مرحله‌ی پنج تا هفت برگی رسیدند، با سوسپانسیون اسپور با غلظت 106 ×1/2 آلوده شدند و در زمان‌های 0، 6، 12، 24، 48، 72 و 96 ساعت پس از مایه‌زنی گیاه با قارچ a. rabiei،rna کل از برگ‌های گیاهان مورد آزمایش استخراج و رشته اول cdna سنتز شد. سطح تظاهر ژن‌های مذکور در نمونه های تیمار شده و شاهد، در ژنوتیپ نخود حساس و مقاوم به برق‌زدگی با استفاده از روش ارزیابی realtime pcr بررسی شد. سطح بیان هر چهار ژن مورد بررسی در این تحقیق در گیاه مقاوم نسبت به بیماری a. rabiei افزایش ‌یافت. نتایج نشان داد که ژن‌های انتخاب شده snakin2 و protein with leucine zipper در ارقام مقاوم در زمان‌ 12 ساعت پس از مایه‌زنی، ژن afpca از خانواده پپتیدهای ضدمیکروبی در زمان‌های اولیه 246 ساعت پس از مایه‌زنی و همچنین ژن pgip از خانواده پروتئین‌های مهارکننده گالاکتوروناز در زمان 48 ساعت پس از مایه‌زنی حداکثر بیان را دارند. به طور کلی نتایج این تحقیق نشان داد که تمام ژن‌های مورد بررسی در این تحقیق در ارتباط با برهمکنش گیاه و بیمارگر بوده و ممکن است باعث کمک به القاء مقاومت در گیاه شوند.

   Gene Expression Analysis of some Resistance Genes in Ascochyta Blight Infected Chickpea (Cicer arietinum)

Abstract    The fungal disease, ascochyta blight, caused by Ascochyta rabiei is a major yieldlimiting factor of chickpea (Cicer arietinum L.) around the world. A clear understanding of the chickpea defense mechanism against A. rabiei is very important for breeding resistant cultivars and better management of this fungal disease. Induced resistance to a pathogen is one of the ways which plants use against biotic stresses. This study was conducted in 2018 as a twofactor factorial experiment (timegenotype) in a completely randomized design with three replications for each of the selected times and the control plant in the laboratory of the Plant Breeding Department of the Faculty of Agriculture, University of Mohaghegh Ardabili. In this study, AFPca, Protein with leucine zipper, Snakin2, and PGIP genes were selected and plant responses to pathogen were studied in two susceptible and resistant genotypes. The experimental system was conducted in a greenhouse, for both inoculated and mockinoculated plants a control plant with three technical replicates. RNA extractions from FLIP0040C (resistant) and FLIP03135C (sensitive) genotypes were performed using RNXplus kit at 0, 6, 12, 24, 48, 72 and 96 hours post inoculation (hpi) and also for mockinoculated plants. The expression of these genes after inoculation by Ascochyta rabiei was measured in susceptible and resistant plants via RealTime PCR. The results showed an increase in the expression level of all genes studied in this research in resistant cultivar compared with susceptible cultivar. Results showed that the candidate gene from the antimicrobial family (AFPca) was upregulated in resistant cultivar at early hours after inoculation (624 hpi), and also for PGIP from galacturonaseinhibit protein family, the gene was maximally expressed to early hours of inoculation to 48 hpi. In general, it can be concluded that all genes studied in this research contribute to plantpathogen interactions and all of them may increase the resistance response to Ascochyta blight disease.
