تعیین اثرات هروئین بر روی ترکیبات کیسه آمنیوتیک در موش های باردار و تخمدان فرزندان آنها

چکیده
|
هروئین یکی از اعتیاد آورترین مواد مخدری است که در حال حاضر در ایران در میان نوجوانان و جوانان مورد سوء مصرف قرار میگیرد. از آنجا که استفاده از مواد مخدر میتواند اثرات زیان باری بر روی اعضاء بدن، بافتها، هورمونها، مایع آمنیوتیک، بیان ژن های موثر در مادر و جنین، و نیز سایر فاکتورها داشته باشد، بنابراین بررسی اثرات هروئین بر کیسه آمنیون موشهای باردار و تاثیرات آن بر نسلهای بعد آن نیز بسیار ضروری میباشد. 60 سر موش صحرایی بالغ ماده نژاد ویستار به 6 گروه کنترل، شم، دریافت کننده دوز 1 میلی گرم/کیلوگرم هروئین، دریافت کننده دوز 2 میلی گرم/کیلوگرم هروئین، دریافت کننده دوز 3 میلی گرم/کیلوگرم هروئین و دریافت کننده دوز 4 میلی گرم/کیلوگرم هروئین تقسیم شدند. در انتها، خونگیری از قلب موشها انجام شد و پارامترهای تستوسترون، گلوکز، lh، fsh، ast، alt، wbcوrbc اندازه گیری شدند. در ادامه، مایع آمنیوتیک جهت سنجش میزان کربوهیدرات ها، فسفولیپید، چربی ها، پروتئین و اوره جدا شد. در پایان، دادهها از نظر آماری مورد ارزیابی قرار گرفتند. تزریق درون صفاقی هروئین تغییرات قابل توجهی را در میزان پرامترهای تستوسترون و تریگلیسیرید نسبت به گروه کنترل ایجاد نمود. این در حالی است که در میزان lh و fsh تغیر قابل توجهی مشاهده نشد. همچنین افزایش بیان ژنهای در نتیجه افزایش دوز هروئین دیده شد. در این بررسی تزریق هروئین حتی در دوزهای پایین توانست باعث افزایش یا کاهش فعالیت تخمدان و اووژنز و در نتیجه اختلال در بلوغ اووسیتها و کیفیت تخمکگذاری گردد.

Effect of Heroein on Amniotic Sac Compounds in Pregnant Rats and Ovarian of Their Female Offspring

Abstract
|
Heroin is one of the most addictive drugs used among adolescents and young people in Iran. Since the use of drugs such as Heroin has detrimental effects on organs, tissues, hormones, amniotic fluid, and expression of genes affecting the mother and fetus, as well as other factors, the aim of this study was to investigate the effects of methamphetamine on the amniotic sac of pregnant rat and its effects on subsequent generations. Sixty adult Wistar female rats were assigned to six groups: control group, sham group, group receiving 1 mg/kg heroin, group receiving 2 mg/kg heroin, group receiving 3 mg/kg heroin and the group receiving 4 mg/kg heroin. At the end of the experimental period, blood samples were taken from the hearts of the mice and the desired parameters such as testosterone, glucose, LH, FSH, AST, ALT, WBC, and RBC were measured. The rats were then dissected; amniotic fluid was isolated to assess the amount of carbohydrates, protein, phospholipids, fats, and urea. Finally, statistical analysis was performed. Intraperitoneal injection of heroin caused significant changes in testosterone and T.G parameters compared to the control group. However, no significant change was observed in LH and FSH levels. In addition, many changes such as weakening and thinning of the theca layer in the graph follicle, destruction of the underlying material and growing follicles following heroin use in the ovarian tissue. Moreover, increased gene expression was seen after increasing the dose of heroin.In this study, heroin injection, even in low doses, could increase or decrease ovarian activity and oogenesis, resulting in impaired oocyte maturation and ovulation quality.
