اثر نوع محیط کشت پایه و تنظیم‌کننده‌های رشد گیاهی بر کشت درون شیشه‌ای Sorbus Persica Hedl.

بارانک ایرانی (sorbus persica hedl.) درختی کُند رشد از تیره رزاسه و از گونه‌های انحصاری ایران و در حال انقراض است. از لحاظ ذخیره ژنی، حفظ زیست محیطی و کاربرد دارویی حائز اهمیت است. به منظور تکثیر درون‌شیشه‌ای این گونه به روش‌ کشت جوانه جانبی، پژوهشی به‌صورت فاکتوریل بر پایه طرح کاملاً تصادفی در سه سطح محیط کشت و سه سطح سیتوکینین در سه تکرار اجرا شد. برای شاخه‌زایی، محیط‌های‌کشت ‌dkw، wpm و ms حاوی سیتوکینین‌های bap، kin و 2ip و برای ریشه‌زایی، محیط‌های کشت ‌dkw، wpm و ms کامل و تغییر یافته با اکسین‌های iba و naa به‌صورت مجزا و کاربرد متوالی بررسی شدند. نتایج نشان داد که بهترین تیمار سترون‌سازی، کاربرد متوالی محلول هیپوکلریت سدیم 20% (حجمی/حجمی، 15 دقیقه) و کلرید جیوه 0.1% (3 دقیقه) در فصل تابستان بود. تکثیر بهینه و رشد طولی شاخساره‌ها در محیط کشت ms با تنظیم‌کننده‌های رشد iba (0.01 میلی‌گرم در لیتر)، kin (0.25 میلی‌گرم در لیتر) و bap (3 میلی‌گرم در لیتر) مشاهده شد. همچنین، ریشه‌زایی بهینه در محیط کشت dkw با یک چهارم غلظت عناصر بیش مغذی حاوی هورمون‌های naa+iba (0.3+0.3 میلی‌گرم در لیتر) بدست آمد.

Effects of culture medium type and plant growth regulators on in vitro culture of Sorbus persica Hedl.

Sorbus persica Hedl. is an Iranian endemic slowgrowing tree (fam. rosaceae) and is endangered. It is important in terms of the gene storage, environmental protection, and medicinal uses. To in vitro propagate this species by the lateral bud culture, a factorial experiment was conducted in a completely randomized design in three levels of culture medium and three levels of cytokinin with three replications. For shooting, the DKW, WPM, and MS culture media containing the cytokinins BAP, Kin, and 2ip, and for rooting, the complete and modified DKW, WPM, and MS culture media containing the auxins NAA and IBA separately and in a consecutive application were investigated. The results showed that the best sterilization treatment was a consecutive application of sodium hypochlorite 20% (v/v, 15 min) and mercuric chloride 0.1% (3 min) in summer. The best proliferation and longitudinal growth of shoots was observed in the MS medium containing IBA (0.01 mg l1), Kin (0.25 mg l1), and BAP (3 mg l1) growth regulators. Also, the best rooting was obtained in the DKW medium with a quarter of the concentration of macroelements containing the NAA+IBA hormones (0.3+0.3 mg l1).
