   اثرات هشت هفته تمرین هوازی بر کاینورنین و بیان ژن و پروتئین گیرنده آریل هیدروکربن بافت قلب موش‌های صحرایی نر

چکیده    هدف: شناخت سازوکارهای سلولی مولکولی قلب به منظور پیشگیری و درمان بیماری‌های قلبی عروقی ضروری به نظر می‌رسد. هدف از پژوهش حاضر بررسی تاثیر تمرین هوازی بر کاینورنین و بیان ژن و پروتئین گیرنده آریل هیدروکربن (ahr) بافت قلب موش‌های صحرایی نر نژاد ویستار بود.مواد و روش‌ها: مطالعه حاضر از نوع تجربی است که تعداد 12 سر موش صحرایی نر نژاد ویستار تهیه و بعد از سازگاری با محیط به 2 گروه: کنترل و تمرین تقسیم شدند. سپس موش‌های صحرایی گروه تمرین به مدت هشت هفته، 5 جلسه در هفته، با سرعت فزاینده بین 10 تا 26 متر بر دقیقه، تمرین دویدن بر روی تردمیل را اجرا کردند. پس از بافت برداری و استخراج rna و تهیه cdna، از روش پی‌سی‌آر جهت اندازه‌گیری بیان ژن آریل هیدروکربن و از روش الایزا برای اندازه‌گیری بیان پروتئین ahr و کاینورنین استفاده شد.یافته‌ها: نتایج آزمون تی مستقل نشان داد که تمرین هوازی منجر به کاهش معنادار بیان پروتئینahr (0.001=p) و متابولیت کاینورین (0.003=p) شده است، اما در سطح بیان ژن ahr تغییر معناداری مشاهده نشد.نتیجه‌گیری: با توجه به تاثیر مثبت تمرین هوازی بر عملکرد قلب و کاهش ahr کاینورنین توسط این نوع تمرین، به نظر می‌رسد کاهش سطح فعالیت این مسیر پیام‌رسانی، برای بهبود عملکرد قلب در شرایط عادی، مناسب است.

   Effects of eight weeks aerobic training on kynurenine and gene and protein expression of aryl hydrocarbon receptor in the heart of male rats

Abstract    Introduction: Understanding the molecular cellular mechanisms of the heart is essential for the prevention and treatment of cardiovascular disease. This study aimed was to investigate the effect of aerobic exercise on the kynurenine and gene and protein expression of aryl hydrocarbon receptor (AHR) in the heart tissue of healthy male Wistar rats.Materials and Methods: The present study is an experimental study design that twelve male Wistar rats were prepared and after adaptation to the environment were divided into 2 groups: control and training. Then, rats in the exercise group performed running on the treadmill for eight weeks, 5 sessions per week, speed between 10 and 26 meters per minute increasingly. After tissue resection and RNA extraction and cDNA preparation, the RealTime PCR method was used to measure the gene expression of Ahr, and the ELIZA method was used to measure the expression of AHR protein and kynurenine.Results: The results showed that aerobic exercise led to a significant decrease in the protein expression of AHR (P=0.001) and metabolite of kynurenine (P=0.003), but no significant changes were observed in the level of Ahr gene expression.Conclusion: Considering the positive effect of aerobic exercise on heart function and the reduction of AHR and kynurenine by this type of exercise, it seems that reducing the activity level of this cell signaling pathway is suitable for improving heart function under normal conditions.
