بررسی اثرات تحریک گیرندههای بتا2-آدرنرژیک در ناحیه قاعدهای-جانبی آمیگدال بر رفتارهای شبه-اضطرابی بهدنبال استرس شوک الکتریکی کف پا در موش صحرایی نر

چکیده
|
هدف: ناحیه قاعدهای-جانبی آمیگدال ساختار بسیار مهم در انسجام اطلاعات اضطراب میباشد. پیشنهاد شده است که ناحیه قاعدهای-جانبی آمیگدال، آورانهای نورآدرنرژیک زیادی از هسته لوکوس سرلئوس دریافت میکند. علاوه بر این، استرس سبب افزایش ترشح نوراپینفرین در ناحیه قاعدهای-جانبی آمیگدال میشود. بنابراین، در مطالعه حاضر، اثرات تحریک گیرندههای بتا2-آدرنرژیک در ناحیه قاعدهای-جانبی آمیگدال بر رفتارهای اضطرابی بهدنبال استرس شوک الکتریکی بررسی میشود.مواد و روشها: استرس شوک الکتریکی (10 هرتز، ولتاژ 40 میلیولت به مدت 60 ثانیه) به کف پای حیوان به مدت 4 روز پشت سر هم اعمال شد. سالبوتامول (آگونیست گیرنده بتا2-آدرنرژیک، μg/rat4) به صورت دوطرفه در ناحیه قاعدهای-جانبی آمیگدال، 5 دقیقه قبل از شروع استرس در هر روز (به مدت 4 روز پشت سر هم) تزریق شد. رفتارهای اضطرابی، 24 ساعت بعد از آخرین روز استرس با استفاده از تستهای ماز مرتفع بعلاوهای شکل و جعبه باز ارزیابی شد.یافتهها: استرس شوک الکتریکی سبب کاهش معنیدار زمان حضور و دفعات ورود به بازوی باز در تست ماز مرتفع شد. به طور مشابه، استرس سبب کاهش معنیدار دفعات ورود و زمان سپری شده در ناحیه مرکزی جعبه باز شد. همچنین، استرس سبب کاهش معنیدار تعداد grooming و rearing شد. تزریق سالبوتامول سبب افزایش معنیدار زمان حضور و نیز دفعات ورود به بازوی باز در مقایسه با گروه استرس شد. همچنین، سالبوتامول سبب افزایش زمان حضور در ناحیه مرکزی، دفعات ورود به ناحیه مرکزی، تعداد grooming و rearing، عبور از خطوط و مسافت کل طی شده شد.نتیجهگیری: گیرندههای بتا دو آدرنرژیک در ناحیه قاعدهای جانبی آمیگدال احتمالاً واکنشهای اضطرابی القا شده ناشی از استرس شوک الکتریکی را واسطهگری میکنند.

Effects of the stimulation of β2-adrenoceptors of the the basolateral amygdala on anxiety-like behaviors following electrical foot-shock stress in male rat

Abstract
|
Introduction: The basolateral amygdala (BLA) is a key structure that integrated anxiety information. It is suggested that basolateral area of amygdala receives a dense norepinephrine innervation from the locus coeruleus nucleus. Furthermore, stress increased norepinephrine secretion in the BLA. In the present study, we examined the effects of the stimulation of beta;2adrenoceptors in the BLA on anxietylike behaviors following footshock stress.Materials and Methods: Footshock stress (10 Hz, 40 mV for 60 seconds) for 4 consecutive days applied to animals. Salbutamol (the beta;2adrenoreceptor agonist; 4 μg/rat) bilaterally injected into the BLA 5 min before starting footshock stress for 4 consecutive days. Anxietylike behaviors assessed 24h after the last session of stress, using elevated plusmaze (EPM) and open field test (OFT).Results: Footshock stress significantly decreased time spent and the number of entries into the open arms of EPM. Similarly, stress significantly decreased the time spent and the number of entries into the center area of OFT. Furthermore, footshock stress significantly decreased the number of grooming and rearing behaviors. Additionally, intraBLA infusions of salbutamol before stress significantly increased time spent and the number of entries into the open arms as compared with stress group. In addition, OFT results showed that salbutamol increased the time spent in the center, number of center entries, the number of rearing and grooming behaviors, line crossing and total distance as compared with the stress group.Conclusion: It is likely that beta;2 adrenoceptors of the BLA mediate the inducedanxietybehaviors by footshock stress.
