   اثر تمرین هوازی فزاینده، تزریق سلول‌های بنیادی مشتق از بافت چربی و ترکیب آنها بر میزان پروتیین Bax و Bcl-2 سلول‌های بتای پانکراس موش‌های دیابتی

چکیده    هدف: آپوپتوز یک فرآیند سلولی محافظت‌کننده است که نقش مهمی در توسعه و هومئوستاز بافت طبیعی و نیز پیدایش بیماری‌ها دارد. مطالعه حاضر با هدف تعیین تاثیر تمرین هوازی فزاینده به همراه تزریق سلول‌های بنیادی مشتق از بافت چربی بر میزان پروتیین bax و bcl2 سلول‌های بتای پانکراس موش‌های صحرایی دیابتی شده با استرپتوزوسین بود.مواد و روش‌ها: در یک مطالعه تجربی، 48 سرموش به شش گروه: کنترل، شم، دیابت، تمرین، سلول درمانی ، تمرین+ سلول درمانی تقسیم شدند. برای ایجاد مدل دیابت، stz با دوز 60 میلی‌گرم در کیلوگرم در ترکیب با بافرسیترات و 4.5= ph به صورت درون صفاقی تزریق شد. گروه تمرینی، شش هفته و هفته‌ای پنج روز با شدت 7060 درصد vo2max روی نوارگردان دویدند. به گروه سلول درمانی، 5 میلی لیتر pbs حاوی 106 ×1.5 عدد سلول استخراج شده از بافت چربی انسانی از طریق سیاهرگ دمی تزریق شد.. مقادیر bax ( به عنوان یک پروتیین پیش برنده مرگ سلولی) و bcl2 ( به عنوان یک پروتیین ضد مرگ سلولی) سلول‌های بتای پانکراس، بعد از هموژنیزوسانتریفیوژ، به وسیله کیت آزمایشگاهی با روش الایزا اندازه‌گیری شد. یافته‌ها: نتایج نشان داد که در گروه دیابتی نسبت به گروه کنترل، سطح bax به میزان معنی داری افزایش و میزانbcl2 به میزان معنی داری کاهش یافت و نسبت bax/bcl2 نیز افزایش یافت (p<0.001). این مقادیر، در گروه های دیابتی با تمرین، سلول درمانی ، تمرین+ سلول درمانی اصلاح شد به گونه ای که تفاوت معنی داری در همه شاخص ها با گروه دیابتی فاقد مداخله وجود داشت (p<0.001).نتیجه‌گیری: به نظر می‌رسد که ورزش، سلول درمانی و استفاده ترکیبی آنها می تواند از آپوپتوز سلولهای بتا جلوگیری نماید.

   Effect of progressive aerobic training, injection of adipose tissue-derived stem cells, and their combination on Bax and Bcl-2 levels of beta - pancreatic cells in diabetic rats

Abstract    Introduction: Apoptosis is protective cellular process the plays an important role in the development and homeostasis of natural tissue as well as diseasecausing factors. Current study was accomplished in order to determine the effect of progressive aerobic training with injection of adipose tissuederived stem cells Bax (as an apoptotic protein) and Bcl2 (as an antiapoptotic protein) and the ratio of Bax to Bcl2 of betapancreatic cells in diabetic rats with streptozotocin.Materials and Methods: 48 rat rsquo;s adult rats were divided into 6 groups: control, sham, diabetes, exercise, stem cell and exercise +stem cell. Diabetes was induced by intraperitoneal injection of streptozotocin (60 mg/kg) dissolved in citrate buffer, with pH = 4.5. The exercise protocol was treadmill running for 5 days per week with intensity of 6070% VO2max for 6 weeks. In the stem cell group, 5 ml PBS with 1.5 ×106 stem cells extracted from human adipose tissue of diabetic rats was injected into the tail vein. Levels of Bax and Bcl2 of beta pancreatic cells were measured by laboratory kit.Results: The findings showed that the levels of Bax and Bcl 2 significanly increased and decreased in the diabetic grop than the control group, respectively (P< 0. 000). Bax/Bcl2 ratio also increased in the diabetic group than the control group. The diabetic groups treated with exercise, stem cells, and both exhibited signifiacly lower Bax and higher Bcl2 levels than the diabetic group alone. Moreover, in these groups Bax/Bcl2 ratio was sigficanly lower than the diabetic group alone.Conclusion: It seems that exercise, stem cell therapy and the combined treatement could prevent beta;cell apoptosis.
