   بررسی اثرات عصاره متانلی گیاه Epilobium Parviflorum بر رشد و عملکرد سلول‌های کبد انسانی Hepg2

چکیده    هدف: عصاره‌های بید علفی پونه‌ای) (epilobium parviflorum ترکیبات آنتی‌اکسیدانت قوی دارد و رشد انواع معدودی سلول‌های سرطانی را مهار می‌کند. هدف این مطالعه بررسی عصاره‌های این گیاه بر رشد سلول‌های کبدی hepg2 و نیز در شرایط تنش اکسیداتیو بود.مواد و روش‌ها: عصاره‌ی متانلی بخش‌های مختلف گیاه جداگانه تهیه شد. زنده‌مانی سلول‌ها پس از تیمار 48 ساعته با عصاره‌ها و به دنبال آن با یا بدون تیمار با h2o2، به روش mtt ارزیابی گردید. ظرفیت آنتی‌اکسیدانی عصاره‌ها و فعالیت ترانس آمینازها، سوپر اکسید دیسموتاز و لیپید پراکسیداز در سلول‌های تیمار شده سنجش شد.یافته‌ها: نه تنها هیچ‌یک از عصاره‌ها سمیتی بر سلول‌های hepg2 نداشتند، بلکه زنده‌مانی سلول‌ها در تیمار با عصاره‌های بخش‌های هوایی و ریشه افزایش معناداری نشان داد (0.006=p). در مدل تنش اکسیداتیو، زنده‌مانی سلول‌های پیش تیمار شده با عصاره‌ها و به ویژه عصاره بخش‌های هوایی افزایش معنی‌دار داشت (p≤0.05). پیش تیمار با این عصاره در سلول‌های تحت تنش به‌طور وابسته به غلظت باعث کاهش mda و فعالیت آنزیم‌های alt و ast درون سلولی و افزایش فعالیت آنزیم سوپر اکسید دیسموتاز در مقایسه با کنترل شد (p≤0.05). ظرفیت آنتی‌اکسیدانی عصاره‌ها قابل مقایسه با ترولوکس برآورد شد.نتیجه‌گیری: عصاره متانلی epilobium parviflorum بر سلول‌های hepg2 سمیت ندارد و احتمالاً می‌تواند سلول‌های کبدی انسان را در برابر تنش‌های اکسیداتیو محافظت کند. لذا می‌تواند پیشنهاد ارزشمندی برای پیشگیری از آسیب‌های کبدی باشد.

   Effects of methanolic extracts of Epilobium parviflorum on growth and function of human liver cells HepG2

Abstract    Introduction: Epilobium parviflorum, the willowherb, extracts contain strong antioxidant compounds and inhibit few types of cancer cell. This study investigates for the first time the effect of its extracts on the growth of human liver cells HepG2, also under oxidative stress.Materials and Methods: Methanolic extract of roots, shoots and flowers was prepared separately. Cell viability was assessed by MTT method after 48 hours of treatments with extracts and then with or without hydrogen peroxide treatment. The antioxidant capacity of the extracts as well as the activities of transaminases, superoxide dismutase and lipid peroxidase were measured in the treated cells.Results: None of the extracts showed cytotoxicty against HepG2 cells, nevertheless cell viability in treatment with aerial and root extracts was significantly increased (P<0.05). In the oxidative stress model, the survival of cells pretreated with extracts, especially methanolic extracts of aerial parts, increased significantly (P<0.05). Pretreatment with this extract in cells under oxidative stress decreased the concentration of MDA and the activity of intracellular ALT and AST enzymes and also increased the activity of superoxide dismutase enzyme compared to the control. The antioxidant capacity of the extracts was comparable to that of Trolux.Conclusion: Methanolic extract of Epilobium parviflorum showed no cytotoxicity on HepG2 cells and probably protect human liver cells against oxidative stress. It can be a valuable suggestion for preventing liver damages.
