   القاء اثرات آپوپتوتیک و ضدتکثیری در سلول‌های Reh متعاقب مهار رسپتور نورکینین-1 توسط Aprepitant

چکیده    هدف: به دنبال معرفی مسیر پیام‌رسانی nk1r به عنوان یک مسیر کارسینوژنیک، مطالعات زیادی، فعالیت غیرطبیعی این مسیر را در بسیاری از بدخیمی‌های انسانی گزارش کرده‌اند. بر اساس اهمیت قابل توجه مسیر nk1r در پاتوژنز لوسمی لنفوبلاستیک حاد (acute lymphoblastic leukemia) (all)، هدف این مطالعه بررسی تاثیر مهارکننده غیرپپتیدی و رقابتی ) nk1r (aprepitant، ا بر رده سلولی reh، مشتق شده از لوسمی لنفوبلاستیک حاد بود.مواد و روش‌ها: برای بررسی اثر آنتی‌لوسمیک آنتاگونیست nk1r، سلول‌های reh با دوزهای مختلفی از aprepitant تیمار گشتند و سپس درصد زنده‌مانی، میزان تکثیر سلولی و فعالیت متابولیک آن‌ها مورد ارزیابی قرار گرفت. علاوه بر این، به منظور مشخص کردن تاثیر دارو بر پیشرفت چرخه سلولی و القاء آپوپتوز، رنگ‌آمیزی annexinv/pi و آنالیز محتوای dna سلولی صورت گرفت. در انتها، میزان تغییر فعالیت رونویسی ژن‌های مرتبط با آپوپتوز توسط تست rqpcr مورد ارزیابی قرار گرفت.یافته‌ها: ما دریافتیم که مهار مسیر پیام‌رسانی nk1r دارای اثرات سایتوتوکسیک و آنتی‌پرولیفراتیو در رده سلولی reh از طریق افزایش خروج فسفاتدیل سرین، افزایش جمعیت سلولی در فاز g1 و کاهش زنده‌مانی سلول‌های تیمار‌شده با aprepitant می‌باشد. علاوه بر این، نتایج ما نشان داد که آپوپتوز القاء‌شده توسط aprepitant در این رده سلولی احتمالاً از طریق تغییر نسبت بیان ژن‌های آنتی‌آپوپتوتیک و پروآپوپتوتیک، bax و bcl-2، صورت می‌گیرد.نتیجه‌گیری: این مطالعه تاثیر ضد لوسمیک aprepitant را در رده سلولی pre-b all آشکار کرده و توانایی این دارو را در القاء آپوپتوز مشخص می‌سازد.

   Induction of apoptotic and anti-proliferative effects in REH cells upon neurokinin-1 receptor inhibition using Aprepitant

Abstract    Introduction: Following the first description of neurokinin1 receptor (NK1R) network as a carcinogenic pathway, overwhelming numbers of studies have reported the inappropriate activity of this cascade in a wide assortment of human cancers. On the basis of the principal importance of NK1R in the pathogenesis of acute lymphoblastic leukemia (ALL), it was of great interest to dissect the effect of Aprepitant, a competing nonpeptide NK1R antagonist, on REH Pre-B ALL cells.Materials and Methods: To evaluate the antileukemic effect of NK1R antagonist, REH cells were exposed to the increasing concentrations of Aprepitant and subsequent cell viability, growth kinetics, and metabolic activity were investigated. Moreover, to underscore whether Aprepitantinduced cytotoxic effect could be attributed to either induction of apoptosis or cell cycle arrest, AnnexinV/PI staining and DNA content analysis were performed. Finally, the transcriptional alterations of apoptosisrelated target genes were studied using RQPCR.Results: Intrestingly, we found that abrogation of the NK1R signaling pathway exerts cytotoxic and antiproliferative effects against REH cells, as evidenced by the increased externalization of phosphatidyl serine (PS), elevated G1 cell population, and decreased number of inhibitortreated viable cells. In addition, our results also suggested that Aprepitantinduced apoptotic cell death in REH cells is mediated, at least partially, through shifting the ratio of death promoters to death repressors via alteration of Bax and Bcl-2 expression.Conclusion: This study highlighted the potent efficacy of Aprepitant in Pre-B ALL cells and shed light on proapoptotic property of this inhibitor.
