   ترمیم سریع زخم پوستی در موش‌های دیابتی با استفاده از نانو داربست پلی کاپرولاکتون - ژلاتین

چکیده    هدف: نانو داربست های سه بعدی با ایجاد محیط مشابه بافت های بدن چشم انداز جدیدی در کشت سلول و ترمیم بافت ایجاد کرده اند، هدف این مطالعه، بررسی اثر داربست نانو فیبر زیست تخریب پلی کاپرولاکتون(pcl) در ترمیم زخم های دیابتی بود. مواد و روش ها: داربست نانو pcl ژلاتین به روش الکتروریسی سنتز شد. در پوست پشت 15 موش آزمایشگاهی زخم پوستی دایره ای به قطر 6 میلی متری ایجاد شد و موش ها به 3 گروه فاقد دیابت (sham)، گروه دیابتی بدون دریافت داربست نانو(control) ، گروه دیابتی دریافت کننده داربست نانو(scaffold) تقسیم شدند. سطح زخم و میزان ترمیم زخم در 3 گروه مورد مطالعه در روزهای7 و 14 بعد از پیوند سلول با استفاده از رنگ آمیزی h e و نرم افزارهای image j وspss بررسی شد. یافته ها: در بررسی سطح زخم در روز 7 کوچک ترین و بیش ترین سطح زخم به ترتیب در گروه scaffold وcontrol مشاهده شد) 45. در روز 14 کوچک ترین و بیش ترین سطح زخم به ترتیب در گروه sham و control مشاهده شد p<0/00. در بررسی درصد بهبودی زخم در روز 7 و 14 بیش ترین درصد بهبودی به ترتیب در گروه scaffold و sham مشاهده شد و در روز 14 تفاوت معنی داری بین این دو گروه وجود نداشت p<0/04. نتیجه گیری: نانوداربست پلی کاپرولاکتون سازگاری مناسبی با زخم های پوستی در موش دارد و می تواند سرعت و میزان ترمیم زخم در موش های دیابتی را افزیش دهد.

   Rapid repair of skin ulcers in diabetic rats using polycaproplactone /gelatin nanon-fiber scaffold

Abstract    Introduction: Threedimensional nano scaffolds by creating a threedimensional structure similar to the body tissues have created a new perspective on cell culture and tissue repair. The aim of this study was to investigate the effect of polycaprolactone biodegradation scaffold (PCL) in treatment of diabetic wounds.Materials and Methods: Nano PCLGelatine scaffold was synthesized by electrospun method. A circular wound (6 mm) was created in skin of 15 BALB/c rats and the rats were divided into 3 groups including: Nondiabetic group (sham), Diabetic group without nano scaffold receiving (control) and diabetic group with nano scaffolds (scaffold). Wound surface area and wound healing were evaluated on the 7th and 14th days using H E staining with Image J and SPSS softwares.Results: On 7th day, the smallest and highest wound surface was observed in the Scaffold and Control groups respectively (P <0.04). On 14th day, the smallest and highest levels of ulcers were observed in sham and control groups respectively (P <0.00). In the evaluation of wound healing on 7th and 14th days, the highest treatment rates were observed in the Scaffold and sham groups respectively, and on the 14th day there was no significant difference between the two groups (0.40).Conclusion: Polycaprolactone nanofiber scaffold has a good compatibility with skin ulcers in rats and it can increase the rate and extent of wound healing in diabetic rats.
