بررسی میزان بیان lncrna dscr9 در بیماران مبتلا به سرطان کولورکتال

زمینه و هدف: بررسی ‌ها نشان داده است که dscr lncrna از سرطان و التهاب جلوگیری می ‌کند و بیان آن منجر به فعالیت‌های ضد توموری می ‌شود. بنابراین هدف از این تحقیق بررسی ارتباط میزان بیان lncrna dscr9 با خطر ابتلا به سرطان روده بزرگ بود.روش تحقیق: از 20 نمونه بافت بیماران مبتلا به سرطان کولورکتال و 20 بافت سالم روده افراد سالم نمونه‌گیری انجام شد. استخراج total rna و سپس سنتز cdna انجام شد. بررسی بیان lncrna dscr9 با استفاده از روش real time rt pcr و پرایمرهای طراحی شده با استفاده از نرم‌افزار beacon designer انجام شد. میزان بیان نسبی lncrna dscr9 محاسبه شد و در نهایت توسط نرم‌ افزار graphpad prism نتایج مورد تحلیل قرار گرفت.یافته‌ها: نتایج نشان داد که بیان dscr9 lncrna در سرطان کولورکتال در مقایسه با گروه کنترل و همچنین در تومور افراد دارای متاستاز و فاقد متاستاز افزایش یافت. از طرفی بیان نسبی  dscr9 lncrna در تومور افراد دارای مرحله 1 و 2 و در افراد دارای تومور با اندازه متفاوت افزایش معناداری را نشان نداد؛ اما بیان نسبی lncrna dscr9 در تومور افراد دارای گرید 1 و 2 افزایش بیان داشت و از نظر آماری معنا‌دار بود (0/0144p=). بررسی اختصاصیت و حساسیت lncrna dscr9 نشان داد که به‌عنوان یک نشانگر (0/0001p=) می‌تواند جمعیت بیمار را از سالم جدا کند.نتیجه‌گیری: در تحقیق حاضر بیان نسبی lncrna dscr9 در افراد مبتلا به سرطان کولورکتال در مقایسه با کنترل افزایش بیان داشت که احتمالاً می تواند به دلیل فعال کردن پروتئین های دخیل در سرطان و افزایش مسیر سیگنالینگ wnt/beta-catenin باشد و باعث رشد سلول ‌های تومور و رگ‌ زایی در سرطان باشد. بنابراین با توجه به تغییرات بیان ژن dscr9 lncrna  در سرطان کولورکتال احتمالاً می ‌توان از این ژن به عنوان یک نشانگر زیستی برای تشخیص تومور استفاده نمود.

study of lncrnadscr9 expression rate in colorectal cancer patients

background and aims: studies have shown that lncrna dscr9 prevents cancer and inflammation and its expression leads to anti-tumor activity. therefore, this study aimed to investigate the relationship between lncrna dscr9 expression and the risk of colon cancer.materials and methods: samples were taken from 20 tissues of patients with colorectal cancer and 20 healthy intestinal tissues of healthy individuals. total rna extraction was performed, followed by cdna synthesis. expression of lncrna dscr9 was investigated using real-time reverse transcription–polymerase chain reaction and primers designed using beacon designer software. the relative expression of lncrna dscr9 was calculated, and finally, the results were analyzed by graphpad prism software.results: the results showed that lncrna dscr9 expression increased in colorectal cancer compared to the control group as well as in tumors of metastatic and non-metastatic individuals. however, the relative expression of lncrna dscr9 in tumors of individuals with stages 1 and 2 and in tumors of with different sizes did not show a significant increase; nevertheless, the relative expression of lncrna dscr9 in tumors of grades 1 and 2 was statistically significant (p=0.0144). the specificity and sensitivity of lncrna dscr9 showed that as a marker (p=0.0001), it could isolate the patient population from the healthy one.conclusion: in the present study, there was a relative increase in the expression of lncrna dscr9 in patients with colorectal cancer compared to control, which possibly could be due to the activation of proteins involved in cancer and increased wnt/beta-catenin signaling pathway and cause tumor cell growth and angiogenesis in cancer. therefore, due to changes in the expression of the lncrna dscr9 gene in colorectal cancer, this gene may be used as a biomarker for the detection of tumors. *corresponding author: noosha zia jahromi; emails: nooshazia.59@gmail.comview orcid id profileyou can also search for this author in:  pubmed     researchgate   scopus    google scholar    google scholar profile
