ارتباط چندشکلی rs6543115 در پروموتر ژن il1rl1 با افزایش خطر ابتلاء به سرطان کولون: یک مطالعه توصیفی

زﻣﯿﻨﻪ و ﻫﺪف: ﯾﮑﯽ از ژنﻫﺎﯾﯽ ﮐﻪ در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﻧﻘﺶ آن ﻣﺸﺨﺺ ﺷﺪه اﺳﺖ، interleukin 1 recptor like il1rl1 1 ﻣﯽﺑﺎﺷﺪ. اﯾﻦ ژن ﮐﺪﮐﻨﻨﺪه ﯾﮏ ﮔﯿﺮﻧﺪه ﺑﺮای اﯾﻨﺘﺮﻟﻮﮐﯿﻦ-33 ﻣﯽﺑﺎﺷﺪ ﮐﻪ ﻫﻤﺮاه ﺑﺎ ﻟﯿﮕﺎﻧﺪ ﺧﻮد در ﺳﺮﻃﺎن ﮐﻮﻟﻮرﮐﺘﺎل ﻧﻘﺶ دارد. ﭼﻨﺪﺷﮑﻠﯽﻫﺎی ﺗﮏ ﻧﻮﮐﻠﺌﻮﺗﯿﺪی ﻣﺘﻌﺪدی در ﺗﻨﻈﯿﻢ ﺑﯿﺎن اﯾﻦ ژن ﻧﻘﺶ دارﻧﺪ ﮐﻪ در اﯾﻦ ﻣﻄﺎﻟﻌﻪ ارﺗﺒﺎط ژﻧﻮﺗﯿﭗﻫﺎی ﭼﻨﺪﺷﮑﻠﯽ rs6543115 در ﭘﺮوﻣﻮﺗﺮ ژنil1rl1 ﺑﺎ ﺳﺮﻃﺎن ﮐﻮﻟﻮن ﻣﻮرد ﺑﺮرﺳﯽ ﻗﺮار ﮔﺮﻓﺘﻪ اﺳﺖ. ﻣﻮاد و روشﻫﺎ: در اﯾﻦ ﭘﮋوﻫﺶ ﺗﻮﺻﯿﻔﯽ، dna ژﻧﻮﻣﯽاز ﻧﻤﻮﻧﻪ ﺧﻮن 100 ﺑﯿﻤﺎر ﻣﺒﺘﻼ ﺑﻪ ﺳﺮﻃﺎن ﮐﻮﻟﻮن و 100 ﻓﺮد ﺳﺎﻟﻢ اﺳﺘﺨﺮاج ﮔﺮدﯾﺪ و ﻧﺎﺣﯿﻪ درﺑﺮﮔﯿﺮﻧﺪه ﭼﻨﺪﺷﮑﻠﯽ ﻣﻮرد ﻧﻈﺮ ﺑﺎ اﺳﺘﻔﺎده از ﭘﺮاﯾﻤﺮﻫﺎی ﻣﻨﺎﺳﺐ و ﺑﺎ ﺗﮑﻨﯿﮏ pcr، اﺑﺘﺪا ﺗﮑﺜﯿﺮ و ﺳﭙﺲ ﺗﻮاﻟﯽﯾﺎﺑﯽ ﺷﺪ و ﻫﻤﻪ ﻧﻤﻮﻧﻪﻫﺎ ﺗﻌﯿﯿﻦ ژﻧﻮﺗﯿﭗ ﺷﺪﻧﺪ. در ﻧﻬﺎﯾﺖ، ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮنﻫﺎی آﻣﺎری ﻣﺠﺬور ﮐﺎی و رﮔﺮﺳﯿﻮن ﻟﺠﺴﺘﯿﮏ ارﺗﺒﺎط ژﻧﻮﺗﯿﭗﻫﺎی rs6543115 در ژنil1rl1 ﺑﺎ ﺳﺮﻃﺎن ﮐﻮﻟﻮن ﺑﺮرﺳﯽ ﺷﺪ. ﯾﺎﻓﺘﻪﻫﺎ: ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﮐﻪ ژﻧﻮﺗﯿﭗ or=0/273 gc، 0/827 - 0/090: ﻓﺎﺻﻠﻪ اﻃﻤﯿﻨﺎن 95%، 0/022=p و or=0/086 cc، 0/232 - 0/032 :ﻓﺎﺻﻠﻪ اﻃﻤﯿﻨﺎن 95%، 0/001

association of rs6543115 single nucleotide polymorphism in the il1rl1 gene promoter with increased risk of colon cancer: a descriptive study

background and objectives: one of the genes that has been proven to play a role in colorectal cancer is il1rl1 interleukin 1 receptor like 1 . this gene encodes a receptor for interleukin 33 that has been shown with its ligand, to be involved in colorectal cancer. several single nucleotide polymorphisms are involved in regulating the expression of il1rl1 gene, so that this study investigated the association between genotypes of rs6543115 polymorphism in the promoter of il1rl1 gene and colon cancer.materials and methods: in this descriptive study, genomic dna was extracted from blood samples of 100 patients with colon cancer and 100 healthy individuals. the region containing the desired polymorphic site was amplified using appropriate primers and pcr technique, and then was sequenced, and all samples were genotyped. finally, logistic regression analysis and chi-square test were used to assess the association between rs6543115 genotypes in il1rl1 gene and colon cancer.results: the results showed that genotype gc or= 0.273, 95%ci: 0.090-0.827, p= 0.022 and cc or= 0.086, 95%ci: 0.032-0.232, p<0.001 as a risk factor in comparison to genotype gg, significantly decreased the susceptibility of colon cancer. also, the results showed that the allele c, in comparison to allele g, significantly reduced the risk of colon cancer or= 0.119, 95%ci: 0.066-0.232, p< 0.001 .conclusion: the rs6543115 polymorphism in the il1rl1 gene appears to be associated with the risk of colon cancer.key words: colon cancer, il1rl1 gene, rs6543115 polymorphism funding: this study was funded by islamic azad university, kazerun branch.conflict of interest: none declaredethical approval: the ethics committee of the islamic azad university, kazerun branch, approved the study ir.iau.kau.rec.1398.013 . how to cite this article: dehbozorgi t, moghanibashi m, mohamadynejad p. association of rs6543115 single nucleotide polymorphism in the il1rl1 gene promoter with increased risk of colon cancer: a descriptive study. j rafsanjan univ med sci 2021; 20 4 : 405-14.
