چکیده
|
زمینه و هدف: پوست پسته غنی از ترکیبات فنلی و یک منبع ارزان قیمت حاوی آنتی اکسیدانها میباشد که میتواند گزینه مناسبی جهت مبارزه با سلولهای سرطانی باشد. این مطالعه با هدف تعیین پتانسیل عصاره هیدروالکلی پوست پسته (pistacia vera) و شکل لیپوزومی آن به عنوان یک عامل احتمالی سمیت سلولی در رده سلولی hepg2 که مربوط به سرطان کبد در انسان میباشد، انجام شد.مواد و روشها: در این مطالعه آزمایشگاهی اثرات مهاری عصاره پوست پسته و شکل لیپوزومی آن بر روی رده سلولهای hepg2 و l929 با استفاده از روش دی متیل تیازول ndash; 2 و 5 دی فنیل تترازولیوم برمید ) 3(4,5dimethylthiazol2yl)2,5diphenyltetrazolium] [(mtt در غلظتهای 0 تا 4 میلیگرم بر میلی لیتر در زمان 24 و 48 ساعت سنجیده شد و در هر رده سلولی، با استفاده از آزمون تعقیبی dunnett، میانگین درصد زندهمانی سلولها مورد تجزیه و تحلیل قرار گرفت.یافتهها: نتایج حاصل نشان داد که عصاره هیدروالکلی پوست پسته باعث کاهش زنده مانی سلولهای hepg2 میشود و با افزایش زمان و افزایش غلظت میزان زنده مانی سلولها کمتر میشود. نتایج فرم لیپوزومی عصاره نیز نشان داد که عصاره لیپوزومی باعث کاهش زنده مانی سلولهای hepg2 می شود. نتیجهگیری: مطالعه حاضر نشان دهنده کندتر آزاد شدن عصاره در شکل لیپوزومی نسبت به عصاره آزاد بود به عبارتی دیگر نشان دهنده آهسته رهش شدن عصاره در فرم لیپوزومی می باشد. همچنین فرم لیپوزومی عصاره اثر سمیت کمتری بر روی سلولهای سالم l929 به نسبت سلولهای hepg2 دارد.

Abstract
|
Background and Objectives: Pistachio skin is rich in phenolic compounds and an inexpensive source of antioxidants, which can be a good option for combating cancer cells. The aim of this study was to determine the potential of pistacia vera hydroalcoholic extract and its liposomal form as a potential cytotoxic agent in HepG2 cell line, which is related to human liver cancer.Materials and Methods: In this in vitro study, the inhibitory effects of pistachio hull extract and its liposomal form on HepG2 and L929 cells were determined by the method of 3 (4, 5dimethylthiazol2yl) 2, 5diphenyltetrazolium (MTT) at the concentrations of 0 to 4 mg / ml in 24 and 48 hrs, and for each cell line, the survival mean of the cells was analyzed using Dunnett rsquo;s test..Results: The results of this study showed that the hydroalcoholic extract of pistachio skin reduced the survival of liver cancer cells (HepG2) and decreased the cell viability by increasing time and concentration. Also, the results of the liposomal form of the extract showed that the liposomal extract reduced the survival of liver cancer cells (HepG2).Conclusion: The findings of this study indicated that the extract was more slowly released in the liposomal form than the free extract.
