افزایش اثرات ضد میکروبی آنتیبیوتیکهای استاندارد در ترکیب با متابولیتهای فعال جدا شده از استرپتومیستهای دریایی: یک مطالعه آزمایشگاهی

چکیده
|
زمینه و هدف: استفاده از مخلوط داروها به عنوان یکی از راههای غلبه بر مشکل مقاومت میکروبی مدنظر است. در این تحقیق اثرات ضدمیکروبی ترکیبات فعال تولید شده توسط استرپتومیستهای دریایی در ترکیب با چند آنتیبیوتیک استاندارد علیه پاتوژنهای مقاوم به دارو بررسی شد.مواد و روش ها: در مطالعه آزمایشگاهی حاضر، قابلیت سویههای جدا شده از دریای مازندران در تولید مواد ضدمیکروبی برعلیه پاتوژنهای مقاوم به دارو با روش دیسک دیفیوژن ارزیابی شد. سپس گونههای تولیدکننده در محیط کشت مایع مناسب کشت داده شدند. آنگاه حداقل غلظت مهارکنندگی (minimum inhibitory concentration; mic) و حداقل غلظت کشندگی (minimum bactericidal concentration; mbc) عصارههای اتیل استاتی با روش میکروبراث دایلوشن بر روی سویههای مقاوم به داروی استافیلوکوکوس اورئوس مقاوم به متی سلین (mrsa) و انتروکوکوس مقاوم به ونکومایسین (vre) تعیین شد. دادهها با استفاده از آزمون t زوجی آنالیز شدند.یافتهها: در غربال گری اولیه، سویههای mn2، mn39، mn40 و mn41 فعالیت ضدمیکروبی وسیعی برعلیه mrsa، vre، سالمونلا تیفی و سودوموناس آئروجینوزا از خود نشان دادند که از بین آنها، بهترین مقدار mic برای ایزوله mn39 با 280 میکروگرم بر میلیلیتر علیه mrsa بهدست آمد. این عصارهها در ترکیب با برخی از آنتیبیوتیکهای استاندارد اثرات افزاینده و در برخی موارد اثرات کاهنده قابل ملاحظهای علیه پاتوژنهای مقاوم به دارو داشتند.نتیجهگیری: ترکیبات فعال تولید شده توسط اکتینومیستها، میتوانند خاصیت ضدمیکروبی قابلتوجهی در ترکیب با آنتیبیوتیکهای استاندارد برعلیه پاتوژنهای مقاوم به دارو داشته باشند که نیازمند بررسیهای بیش تر در پژوهشهای آینده می باشد.

An Increase in Antimicrobial Effects of Standard Antibiotics in Combination with the Active Metabolites Isolated from Marine Streptomyces: A Laboratory Study

Abstract
|
Background and Objectives: Combination therapy has been considered as a potential approach to overcome antimicrobial resistance. In this study the antimicrobial effects of active compounds produced by some marine Streptomyces spp. in combination with some standard antibiotics against multidrugresistant pathogens was investigated.Materials and Methods: In this laboratory study, the bacteria isolated from Caspian Sea were tested for their ability to produce antimicrobial compounds active against some drugresistant pathogens using disc diffusion methods. Then, the producer strains were cultivated in suitable liquid media. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the ethyl acetate extracts were measured against some drugresistant species including methicillinresistant Staphylococcus aureus (MRSA) and vancomycinresistant Enterococcus (VRE) using microbroth dilution technique. Data was analysed using paired ttest.Results: In the first screening, strains MN2, MN39, MN40 and MN41 showed a broad antimicrobial activity against MRSA, VRE, Salmonella typhi and pseudomonas aeruginosa, among which, the best MIC against MRSA was obtained from strain MN39 at 280 ( micro;g/ml). These extracts in combination with some standard antibiotics resulted in a noticeable improved antimicrobial activity against multidrugresistant pathogens, while in some cases the activity was decreased.Conclusion: The bioactive compounds produced by actinomyces possess remarkable antimicrobial properties against drugresistant pathogens when used in a combination therapy with standard antibiotics which requires further research in the future.Key words: Marine Streptomyces, Multidrugresistant pathogens, Antimicrobial effects Funding: This study did not have any funds.Conflict of interest: None declared.Ethical approval: The Ethics Committee of Mashhad University of Medical Sciences approved the study (IR.MUMS.REC.1394.556).
