ﺳﺎﺑﻘﻪ و ﻫﺪف: ﮐﻨﺘﺮل ﻫﯿﭙﺮﮔﻠﯿﺴﻤﯽ ﺑﻌﺪ از ﻏﺬا ﯾﮏ اﺳﺘﺮاﺗﮋی ﻣﻬﻢ در ﻣﺪﯾﺮﯾﺖ دﯾﺎﺑﺖ ﻧﻮع دو و ﮐﺎﻫﺶ ﻋﻮارض ﻣﺰﻣﻦ ﻣﺮﺗﺒﻂ ﺑﺎ آن اﺳﺖ. ﺑﻨﺎﺑﺮاﯾﻦ ﻣﻬﺎر ﮐﻨﻨﺪهﻫﺎی آﻧﺰﯾﻢﻫﺎی ﺗﺠﺰﯾﻪ ﮐﻨﻨﺪه ﮐﺮﺑﻮﻫﯿﺪرات ﻣﺎﻧﻨﺪ آﻟﻔﺎ-ﮔﻠﻮﮐﻮزﯾﺪاز ﻣﯽﺗﻮاﻧﺪ در درﻣﺎن اﯾﻦ ﺑﯿﻤﺎری ﻣﻔﯿﺪ ﺑﺎﺷﻨﺪ. ﻫﺪف از اﯾﻦ ﻣﻄﺎﻟﻌﻪ ﺑﺮرﺳﯽ ﻓﻌﺎﻟﯿﺖ ﺑﺎزدارﻧﺪﮔﯽ آﻟﻔﺎ-ﮔﻠﻮﮐﻮزﯾﺪاز ﺑﺮﺧﯽ از داروﻫﺎی ﺳﻨﺘﯽ در ﺳﻠﯿﻤﺎﻧﯿﻪ ﻣﯽﺑﺎﺷﺪ. ﻣﻮاد و روشﻫﺎ: ﻫﺸﺖ ﮔﻮﻧﻪ ﮔﯿﺎﻫﯽ ﺑﺎ ﻣﺸﺎوره از ادوﯾﻪایﻫﺎی ﻣﻌﺘﺒﺮ در ﺳﻠﯿﻤﺎﻧﯿﻪ ﺗﻬﯿﻪ ﺷﺪﻧﺪ. ﭘﺲ از ﺧﺸﮏ ﺷﺪن ﻧﻤﻮﻧﻪﻫﺎ در ﻫﻮا و ﺗﻬﯿﻪ ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﯽ آﻧﻬﺎ، ﺳﻨﺠﺶ آﻧﺰﯾﻤﯽ ﺑﻪ روش ﻣﯿﮑﺮوﭘﻠﯿﺖ در ﺣﻀﻮر ﭼﻬﺎر ﻏﻠﻈﺖ ﻣﺨﺘﻠﻒ 0/01 ،0/1 ،1 و 0/001 ﻣﯿﻠﯽﮔﺮم در ﻣﯿﻠﯽﻟﯿﺘﺮ از ﻋﺼﺎرهﻫﺎ و ﮐﻨﺘﺮل ﻣﻨﻔﯽ و ﻣﺜﺒﺖ اﻧﺠﺎم ﺷﺪ. در ﻧﻬﺎﯾﺖ آﻧﺎﻟﯿﺰ ﺳﻨﺘﯿﮏ آﻧﺰﯾﻤﯽ و ﺗﻌﯿﯿﻦ ﻧﻮع ﻣﻬﺎر ﺑﺮ روی ﻋﺼﺎرهﻫﺎی ﻣﻮﺛﺮ اﻧﺠﺎم ﺷﺪ. ﯾﺎﻓﺘﻪﻫﺎ: در ﻣﯿﺎن ﯾﺎزده ﮔﻮﻧﻪ ﮔﯿﺎﻫﯽ ﻣﻮرد ﻣﻄﺎﻟﻌﻪ، ﻋﺼﺎرهﻫﺎی ﻣﺘﺎﻧﻮﻟﯽ rheum ribes r. ،rubus idaeus l. وsalix alba l. در ﻏﻠﻈﺖ ﯾﮏ ﻣﯿﻠﯽﮔﺮم ﺑﺮ ﻣﯿﻠﯽﻟﯿﺘﺮ، ﺑﻪ ﺗﺮﺗﯿﺐ ﺑﺎ 98/13±1/05 % ic50=0/085 %93/39±2/11 ، ic50=0/046 ، و 84/46±1/05% ic50=0/125 ، ﺑﯿﺸﺘﺮﯾﻦ ﻓﻌﺎﻟﯿﺖ ﺑﺎزدارﻧﺪﮔﯽ را ﻧﺴﺒﺖ ﺑﻪ آﻟﻔﺎ-ﮔﻠﻮﮐﻮزﯾﺪاز از ﺧﻮد ﻧﺸﺎن دادﻧﺪ. در آﻧﺎﻟﯿﺰ ﺳﻨﺘﯿﮑﯽ ﻣﻬﺎر آﻟﻔﺎ-ﮔﻠﻮﮐﻮزﯾﺪاز، ﻋﺼﺎرهﻫﺎی rheum ribes r. ،rubus idaeus l. ﻣﻬﺎر از ﻧﻮع ﻣﺮﮐﺐ رﻗﺎﺑﺘﯽ- ﻏﯿﺮ رﻗﺎﺑﺘﯽ ، و ﻋﺼﺎره salix alba l. ﻣﻬﺎر از ﻧﻮع ﻏﯿﺮ رﻗﺎﺑﺘﯽ را ﻧﺸﺎن دادﻧﺪ. ﻧﺘﯿﺠﻪﮔﯿﺮی: ﻧﺘﺎﯾﺞ ﻣﻄﺎﻟﻌﻪ ﻧﺸﺎن داد ﮐﻪ ﻋﺼﺎره ﺑﺮگ دو ﮔﯿﺎهsalix alba l. ،rubus idaeus l. و ﻋﺼﺎره رﯾﺸﻪ rheum ribes r. ﻣﯽﺗﻮاﻧﻨﺪ ﭘﺲ از اﻧﺠﺎم ﺗﺴﺖﻫﺎی ﮐﻠﯿﻨﯿﮑﯽ ﻣﻨﺎﺳﺐ، در ﺟﻠﻮﮔﯿﺮی از اﻓﺰاﯾﺶ ﻗﻨﺪ ﺧﻮن ﺑﻌﺪ از ﺻﺮف ﻏﺬا اﺳﺘﻔﺎده ﺷﻮﻧﺪ.

Abstract
|
background and objective: control of hyperglycemia after meals is an important strategy in the management of type ii diabetes, and the reduction of chronic complications associated with it. therefore, inhibitors of carbohydrate-degrading enzymes such as α-glucosidase can be useful in the treatment of this disease. the aim of this study was to evaluate the α-glucosidase inhibitory activity in some traditional medicines in sulaymaniyah province of iraq.methods: eight plant species were prepared with advisory from authentic spiceries in sulaymaniyah. after air drying and preparation of their methanolic extracts, enzyme microplate assay was conducted on them in four different extract concentrations 1, 0.1, 0.01 and 0.001 mg/ml along with negative and positive controls. finally, enzyme kinetic analysis was performed on effective extracts.findings: among the eleven studied plant species, rubus idaeus l., rheum ribes r. and salix alba l. extracts at 1 mg/ml concentration showed highest inhibitory activity 98.13±1.05% ic50=0.046 mg/ml , ic50=0.046 mg/ml ic50=0.085 mg/ml and 84.46±1.12% ic50=0.125 mg/ml , respectively against α-glucosidase,. after kinetic analysis of α-glucosidase inhibition, rubus idaeus l., rheum ribes r. extracts showed mixed type of inhibition competitive- uncompetitive while extract of salix alba showed uncompetitive type of inhibition.conclusion: the leave extract from both rubus idaeus, salix alba and root extract of rheum ribes are remarkable α-glucosidase inhibitors, and may be used in the treatment of type ii diabetics after clinical tests.
