چکیده
|
تناوب باردهی، به مفهوم نوسان شدید در عملکرد سالیانه، باعث ایجاد نوعی تنش غیرزیستی در درختان زیتون در سال پرمحصول میشود. بنابراین میتوان فرضیه تغییر در ظرفیت آنتیاکسیدانی برگ گیاه را در ارتباط با این پدیده ارزیابی کرد. هدف از بررسی حاضر، آزمون این فرض در درختان 12 ساله آور و نیآور زیتون رقم کنسروالیا در زمان گلانگیزی و در طی دو سال، از راه بررسی الگوی فعالیت و بیان ژنهای برخی از آنزیمهای آنتیاکسیدان بود. از بین متغیرهای ارزیابی شده مانند ظرفیت آنتیاکسیدانی، مقدار فنول، فلاونوئید و پروتئین کل، تنها فنول کل بهصورت معنیداری در درختان نیآور بیشتر از درختان آور بود. از بین آنزیمهای پراکسیداز (pod)، سوپراکسیددیسموتاز (sod)، فنیلآلانینآمونیالیاز (pal) و پلیفنولاکسیداز (ppo)، فعالیت های سه آنزیم اول تفاوت معنیدار نشان دادند. مقدار فعالیت آنزیم pod در درختان آور نسبت به نیآور و آنزیم های sod و pal در درختان نیآور نسبت به آور فعالیت بیشتری داشتند. بیان نسبی ژنهای مربوط به آنزیم های pod و ppo در درختان آور و sod و pal در درختان نیآور به صورت معنیداری بیشتر بود. کاهش میزان فعالیت و بیان ژن(های) آنزیم sod در درختان آور، با افزایش این شاخصها در مورد آنزیم pod همرا بود. در کل ساخت ترکیبهای فنولی در درختان نیآور و متابولیسم و مصرف آن در درختان آور زیادتر بود.

Changes in Gene Expression and Activity Pattern of Some Antioxidant Enzymes in Olive Trees (Olea europaea L. cv. Conservalia) in Relation to Alternate Bearing

Abstract
|
The alternate bearing, severe fluctuation in annual fruit yield, causes a type of abiotic stress in olive trees during ONyear, i.e. the year with high fruit load. Hence, one could assume a hypothesis in variation of leaves antioxidant capacity in this regard. The current study was conducted aimed at assaying this hypothesis through analyzing some antioxidant enzymes activities and their gene expression patterns. For this, leaves samples were collected from 12yearold olive (cv. Conservalia) trees, both ON and OFFtype, at flower bud induction phase during two successive years. Among studied criteria such as antioxidant capacity, phenol, flavonoid and total protein contents, the amount of phenol in OFFtrees was substantially higher than ONtrees. The activity of peroxidase (POD), superoxide dismutase (SOD) and phenylalanine ammonialyase (PAL) was considerably influenced by bearing status. In comparison to OFFtrees, the activity of the POD enzyme was higher in ONtrees, and the opposite trend was revealed in terms of SOD and PAL activities. The relative gene(s) expression of POD and polyphenol oxidase (PPO) in ONtrees, and SOD and PAL in OFFtrees was significantly high. While the SOD showed downward trends, the POD witnessed an upward pattern in terms of its activity and gene(s) expression profile in ONtrees. In general, the phenolic compounds synthesis and their consumption are the dominant metabolic process in OFF and ONtrees, respectively.
