کاهش بیان mir-199b در بیماران مبتلا به سرطان لوکمی میلوئیدی حاد (aml)

سابقه و هدف: سرطان خون میلوئید حاد aml) (acute myeloid leukemia، دومین سرطان شایع در ایران و پنجمین سرطان در دنیا است که حدود 8%از کل سرطان ها را به خود اختصاص داده است. میکرو rna ها (mirna)گروه کوچکی از rna کوچک غیر کد شونده هستند که در فرآیندهای مهم سلولی از جمله تکامل و تمایز از طریق تنظیم پس از رونویسی نقش اساسی ایفا می کنند و بیان بسیاری از ژن های هدف را دستخوش تغییر قرار داده و در ایجاد سرطان نقش دارند. هدف از این مطالعه، بررسی بیان mir-199b در بیماران مبتلا به aml می باشد. مواد و روش ها: برای این منظور، در این مطالعه casecontrol، بیان mir-199b در30 نمونه خون بیمار مراجعه کننده به بیمارستان میرزاکوچک خان جنگلی بررسی و با همان تعداد نمونه سالم به عنوان کنترل با روش real time pcr مقایسه گردید. یافته ها: سطح بیان mir-199b در بیماران مبتلا به aml کاهش چشمگیری نشان داد که از لحاظ آماری نیز معنی دار بود (0.001=p). در حالیکه ارتباط معناداری بین جنسیت ، سن بیماران، شمارش گلبول سفید، پلاکت، هموگلوبین، بلاست و دسته بندی fab با بیان این ژن ها مشاهده نشده است (p<0/05). نتیجه گیری: بیان mir-199b به عنوان یک تومور ساپروسور کاهش نشان داده است. بنابراین، بررسی میزان بیان این ژن می تواند به عنوان عامل تشخیصی و همچنین در تعیین پیش آگهی بیماران مبتلا به aml بکار رود.

Down regulation of MiR199b expression in Patients with Acute Myeloid Leukemia (AML)

Background and Aim: Acute Myeloid Leukemia (AML) is the second most prevalent type in Iran and the fifth cancer in the world which includes 8% of the cancers. MicroRNAs (miRNAs) are a class of small noncoding RNAs which have been recently shown to play a crucial role in major cellular processes such as development and differentiation which can change the genes regulation that may lead to cancer. The purpose of this study was to determine the miR199b expression in patients with AML. Materials and Methods: For this purpose, in this casecontrol study, miR199b expression was investigated in 30 patients with AML admitted to Mirzakochak khan Jangali Hospital, Tehran and compared with the same number o healthy individuals as control using Real time PCR. Result: There was a reduction in miR199b expression in patients compared with control group and statistically was significant (P=0.0001(. While, no significant correlation was observed between age, sex, WBC, PLT, HGB, Blast and FAB subtype with the expression of Mir199b (P> 0.05). Conclusion: Mir199b as a tumor suppressor showed a reduction in its expression. Therefore, evaluation of this gene expression, can be used as diagnostic agents and also a prognostic factor in patients with AML.
