بررسی مقدماتی توانمندی نوار پروژسترون گاوی (rapid p4) در سنجش نیمه کمی پروژسترون در فاز فولیکولی سگ ماده

زمینۀ مطالعه:تعیین زمان تخمک گذاری  با استفاده از اندازه گیری پروژسترون برای انجام تلقیح مصنوعی یا جفت گیری در زمان مناسب حائز اهمیت است. هدف: ارزیابی توانمندی نوارهای پروژسترون گاوی در تعیین میزان نیمه کمی پروژسترون در سرم خون سگ‏های ماده است. روش‌کار: این مطالعه روی 5 قلاده سگ نژاد مخلوط، سالم که در فاز آنستروس سیکل جنسی قرار داشتند انجام شد. از داروی کابرگولین برای القاء فحلی استفاده شد. بعد از ورود به فاز پرواستروس از روز 7 پرواستروس تا 2 روز پس از تخمک‌گذاری به صورت روزانه اسمیر واژن، اولتراسونوگرافی و خونگیری انجام شد. سرم نمونه‌های خون به دو قسمت تقسیم شد. یک قسمت برای اندازه‌گیری پروژسترون با روش رادیوایمنواسی (ria) و قسمت دیگر با نوارهای پروژسترون گاوی (rapid p4) برای ارزیابی نیمه کمی پروژسترون. تغییرات تخمدان‌ها بوسیله اولتراسونوگرافی ارزیابی شد و تغییرات چرخه فحلی بوسیله آزمایش سایتولوژی واژن انجام گرفت. نتایج: با آزمون اسپیرمن همبستگی مثبت معنی‌داری بین اندازه‌گیری پروژسترون خون با دو روش ria و نوار مشاهده گردید (0.00=p ) (0.916=r). نتایج نمودار roc و آنالیز کاپا نشان داد که بالاترین میزان توافق بین نوارها و روش ria در سطوح 2 و 5 نانوگرم در میلی لیتر پروژسترون سرم خون است. حساسیت و ویژگی نوار در سطح 5 پروژسترون به ترتیب 88.5 و 93.7 درصد و در سطح 10 پروژسترون 82.4 و 96 درصد است. در پرواستروس اسکورهای32 و تنها در یک سگ اسکور 4 مشاهده گردید. در استروس در دو سگ اسکورهای 5 4 و در سه سگ اسکورهای 5 3 مشاهده گردید. نتیجه‌‌گیری نهایی: نوار‏های پروژسترون گاوی در سنجش نیمه کمی پروژسترون در مراحل سیکل جنسی سگ‏های ماده قابل استفاده هستند. اسکور‏های 4 و 5 نوار‏های پروژسترونی می‌توانند زمان تقریبی تخمک‌گذاری و زمان مناسب جفت‌گیری را تعیین کنند.

Evaluation of Bovine Progesterone SemiQuantitative Test Kit (Rapid p4) for Estimation of Blood Serum Progesterone Levels in Follicular Phase of Bitch

BACKGROUND: Detection of ovulation time using progesterone measurement is important to inseminate or breeding during the optimum time. OBJECTIVES: The aim of present study was evaluation of bovine progesterone semiquantitative test kit for estimation of blood serum progesterone levels in bitch. METHODS: Five healthy intact anestrus bitches were used in the present study. Dogs were treated with cabergolin until onset of proestrus. Vaginal cytology, ultrasonography and blood sampling were performed from the 7th day of proestrus until two days after ovulation. Blood serum samples were divided as follows: one part to measure progesterone level by using RIA, and the other part was investigated by bovine progesterone semiquantitative test kit. Ovarian changes were evaluated by ultrasonography and assessment of the estrus cycle was performed by vaginal cytology examination. RESULTS: Following Spearman analysis, significant positive correlation was observed between the semiquantitative test kit and RIA results (r=0.916; p < /em>=0.00). The results of ROC and Kappa analysis suggested that the highest diagnostic accuracy of progesterone semiquantitative test kit was observed in the blood serum progesterone levels of 2 and 5 ng/ml. The sensitivity and specificity of the used kit at the level of 5 ng/ml of progesterone were 88.5 and 93.7 %, respectively. The sensitivity and specificity of the used kit at the level of 10 ng/ml of progesterone were 82.4 and 96 % and at the level of 10 ng/ml were 82.4 and 96%, respectively. The observed scores during the late proestrus were 23 and just one dog showed score 4. During the estrus phase, in two dogs scores 45 and in three dogs scores 35 were revealed.   CONCLUSIONS: Bovine progesterone semiquantitative test kit is useful for estimation of estrus cycles in bitch. Scores of 4 and 5 of semiquantitative test kit indicate ovulation time and proper breeding time.
