   ردیابی ویروئیدهای مرکبات با روش استخراج اسید نوکلئیک توسط Cf11

چکیده    بیماری‌های کاچکسیا ناشی از ویروئیدcitrus cachexia viroid, ccavd  و اگزوکورتیس ناشی از ویروئید citrus exocortis viroid, cevd مهم‌ترین بیماری‌های ویروئیدی مرکبات در جهان هستند. ویروئیدها با روش‌های متداول سرولوژیکی قابل شناسایی نبوده و روش‌‌های فعلی تشخیص آنها عموماً وقت‌گیر، پر هزینه و با محدودیت‌‌هایی همراه است. در این بررسی امکان خالص‌سازی و شناسایی دقیق این ویروئیدها با استفاده از ستون کروماتوگرافیcf11 بررسی شد. طی سال‌های1390 تا 1392، از سرشاخه‌های22 درخت نارنگی و 15 درخت پرتقال به ترتیب واجد علائم بیماری‌های کاچکسیا و اگزوکورتیس در شرق مازندران نمونه برداری شد. بافت برگ پس از پودر شدن با بافر استخراج مخلوط شد. خالص‌سازی در ستون حاوی پودر سلولز انجام و با افزودن اتانول، مولکول‌های آر ان ای دو رشته‌ای استخراج و توسط استات سدیم رسوب داده شد. رسوب حاصل در آگارز یک درصد الکتروفورز و دو باند اختصاصی در محدوده‌های 300 تا 400 جفت باز (فرم خطی) و7800 7000 جفت باز (فرم حلقوی) مرتبط با این دو ویروئید به دست آمد که به ترتیب در ماه‌های گرم و در تمامی ماه‌های سال با این روش قابل‌شناسایی بود. ماهیت rna دورشته‌ای باندهای به ‌دست آمده، با عدم تخریب آنها توسط آنزیم ‌dnase وهمچنین rnase در حضور محلول نمک طعام 0.3 مولار و حلالیت آن‌ها در محلول licl تایید شد. ماهیت ویروئیدی باندهای به دست آمده توسط واکنش زنجیره‌ای پلی مراز (rtpcr) با استفاده از آغازگرهای اختصاصی cevd و ccavd تعیین گردید. تعداد 500 نهال پرتقال و 300 نهال نارنگی در آزمون سلامت نهال با این روش بررسی شد.با توجه به تکرار پذیری و قابل اعتماد بودن این روش و تایید ماهیت باندهای به دست آمده با آغازگر اختصاصی هر یک از این دو ویروئید در واکنش زنجیره ای پلی‌مراز، این روش برای تشخیص ویروئیدهای مرکبات در برنامه‌های کاربردی تولید نهال سالم با صرف حداقل زمان و هزینه پیشنهاد می‌گردد.

   Detection of Citrus Viroids by CF11 Nucleic Acid Extraction Method

Abstract    Cachexia (Citrus cachexia viroid, CCaVd) and exocortis (Citrus exocortis viroid, CEVd) are the most important citrus viroid diseases in the world. Detection of the viroids through the common serological methods is not feasible and the current methods are time consuming and costly with some limitation. Purification and accurate detection of the viroids was studied by column chromatography with CF11 cellulose powder in presence of ethanol (common method for viruses’ dsRNA detection) in comparison to the conventional methods. During 20102012, sampling was done from twigs of 22 Mandarin and 15 orange trees have symptoms of cachexia and exocortis, respectively, in East Mazandaran. Leaf samples were ground and mixed with extraction buffer. Purification carried out in CF11 column and viroid molecules extracted with ethanol and precipitated by adding sodium acetate. The nucleic acid extracts were analyzed by 1% agarose gel electrophoresis and specific bands of the two viroids with molecular sizes of 70007800bp (circular form) and 300400bp (linear form) were detected in warm and all months, respectively. The dsRNA nature of the bands was confirmed by nuclease treatment (DNaseI and RNaseA) and 2M LiCl extraction methods.Viroid entity of the each nucleic acid sample was recognized by RTPCR method using specific primers of cachexia and exocortis viroids. 500 orange and 300 mandarin seedlings were tested in healthy citrus seedling production program.As regards of advantages of this method (repeatability, reliability, saving time and price) and confirmation by RTPCR, the method is recommended for viroid detection in healthy citrus seedling production programs.
