   بررسی و شناسایی ژنهای کروم سنسینگ در سویه های سودوموناس آئروژینوزا جداشده از نمونه های بالینی انسانی به روش Multiplex Pcr و تعیین مقاومت آنتی بیوتیکی

چکیده    مقدمه: سودوموناس آئروژینوزا یک پاتوژن فرصت طلب و عامل 10 الی 15 درصد از عفونت های بیمارستانی می باشد. وجود ژن های حدت یکی از مهم ترین مکانیسم های تهاجمی سودوموناس آئروژینوزا می باشد و این موضوع از نظر پزشکی از اهمیت ویژه ای برخوردار است. بیان بسیاری از ژن های بیماریزای باکتری سودوموناس آئروژینوزا به وسیله یک سیستم ژنی به نام سیستم quorum sensing (qs) کنترل و تنظیم می گردند. کروم سنسینگ سیستم ارتباطی سلول به سلول با استفاده از مولکول های کوچک sms در ارگانیسم های تک سلولی است. هدف از تحقیق بررسی شیوع ژن های دخیل در سیستم کروم سنسینگ در سویه های سودوموناس آئروژینوزا جداشده از منابع انسانی می باشد.مواد و روش ها: در این مطالعه، 60 نمونه سودوموناس آئروژینوزا از نمونه های گوارش انسانی تهیه و در محیط های اختصاصی کشت و توسط تست های تشخیصی و افتراقی تایید گردیدند. آزمون multiplex pcr برای ردیابی ژن های مورد نظر انجام گرفت. آزمون حساسیت آنتی بیوتیکی علیه 10 عامل ضد میکروبی صورت گرفت.یافته ها: نتایج multiplex pcr نشان داد که میزان فراوانی ژن های rhlr 5 درصد، ژنlasr 3/48 درصد و ژنlasi 60 درصد بوده، در صورتی که ژن های lasb،apr ، rhlab و rhli در هیچ یک از نمونه ها شناسایی نگردیدند. در آزمون حساسیت آنتی بیوتیکی، بیشترین میزان مقاومت نسبت به آنتی بیوتیک های آمیکاسین، آموکسی سیلین و سفوتاکسیم و بیشترین میزان حساسیت به آنتی بیوتیک های سیپروفلوکساسین و سفتازیدیم گزارش گردیده است.بحث و نتیجه گیری: ژن های سیستم qs فراوانی بالایی در بین سویه های سودوموناس آئروژینوزا جداشده از منابع انسانی دارند.

   The study and identification of Quorum Sensing (QS)‎‏ ‏genes of Pseudomonas ‎aeruginosa strains isolated from human clinical samples by Multiplex ‎PCR and antibiotic resistance determination

Abstract    Background : Pseudomonas aeruginosa is an opportunistic pathogen and the cause of 10% to 15% of amp;lrm;nosocomial infections.Virulence genes of Pseudomonas aeruginosa is one of the most amp;lrm;aggressive mechanisms and the issue of medical opinion is important. The expression of many amp;lrm;genes is controlled and regulated in pathogenic bacteria Pseudomonas aeruginosa gene by a amp;lrm;system called Quorum Sensing (QS). QS controls and regulates celltocell communication amp;lrm;system using small molecules in singlecelled organisms SMs. This study was aimed at amp;lrm;defining the prevalence of QS genes in Pseudomonas aeruginosa isolated from human amp;lrm;resources.amp;lrm;Materials and Methods: Initially 60 samples of Pseudomonas aeruginosa from human intestinal samples were prepared amp;lrm;and confirmed by culturespecific diagnostic tests. Multiplex PCR test was amp;lrm;performed to detect genes. Antimicrobial susceptibility of the isolates against 10 antimicrobial agents was amp;lrm;determined using standard disk diffusion method.amp;lrm;Results: Multiplex PCR results showed that the frequency of the desired genes rhlR 5 %, lasR 48.3% and lasI 60%. The genes lasB, apr, rhlAB and rhlI were not detected in any of the samples. According to antibiogram, the most resistance was against amikacin, amoxicillin and cefotaxime and the most susceptibility was to ciprofloxacin and ceftazidime.Conclusion: This study shows high prevalence of Quorom Sensing genes in Pseudomonas aeruginosa isolates of human origin.
