   تعیین شیوع و گونه پلاسمودیوم در پشه های حامل شهرستان نیک شهر در جنوب استان سیستان و بلوچستان توسط روش Multiplex Nested-Pcr

چکیده    مقدمه: مالاریا از جمله مهمترین بیماری های انگلی است که به عنوان یکی از مسایل مهم بهداشتی در کشورهای گرمسیر و نیمه گرمسیر مطرح می باشد. این مطالعه با هدف بررسی میزان و نوع آلودگی پشه های آنوفل ماده به انگل مالاریا با استفاده از روش مولکولی multiplex nested-pcr در جنوب استان سیستان و بلوچستان انجام گردید.مواد و روش ها: در نیمه دوم سال 1396 تعداد 400 پشه آنوفل ماده توسط ایستگاه های چک حشره شناسی علوم پزشکی زاهدان از روستاهای شهرستان نیک شهر جمع آوری گردید. صید پشه ها با استفاده از روش های صید دستی از اماکن داخلی (انسانی و حیوانی) و اماکن خارجی طبیعی و مصنوعی (شلترپیت) انجام شد. پس از استخراج dna، بررسی مولکولی با روش multiplex nested-pcr انجام گردید.یافته ها: از 310 نمونه جمع آوری شده از بخش های لاشار، اهوران و مرکزی شهرستان نیک شهر تعداد 6 نمونه (1.5 %) آلوده به پلاسمودیوم ویواکس و از 90 نمونه جمع آوری شده مربوط به بخش های فنوج و بنت شهرستان مذکور هیچ نمونه مثبتی گزارش نگردید. در این بررسی آلودگی به پلاسمودیوم فالسیپاروم و میکس (پلاسمودیوم ویواکس و پلاسمودیوم فالسی پاروم) مشاهده نشد.بحث و نتیجه گیری: نتایج نشان می دهد که در مکان های که در آن انتقال هر دو گونه پلاسمودیوم ویواکس و پلاسمودیوم فالسی پاروم رخ می دهد، تشخیص انگل های مالاریا به وسیله pcr می تواند یک مکمل بسیار مفید برای تایید تشخیص میکروسکوپی باشد.

   Determination of the prevalence and type of plasmodium in mosquitoes present in the city of Nik Shahr, Southern Sistan and Baluchestan Province, IR Iran, by Multiplex Nested-PCR

Abstract    Background: Malaria is one of the most important parasitic diseases, and is considered one of the important health issues in tropical and subtropical countries. The aim of this study was to determine the rate and type of infection of Anopheles mosquitoes by malarial parasites using Multiplex NestedPCR in the South of Sistan and Baluchestan province.Materials and Methods: In the second half of 2017, 400 Anopheles mosquitoes were collected from Zahedan Medical Insecticide Check Centers located in villages around Nikshahr city. The mosquitoes were caught by handheld methods in domestic (human and animal), natural and artificial outdoor places (Shelterpit). After DNA extraction, molecular analysis was performed using Multiplex NestedPCR.Results: Of the 310 samples collected from parts of Lashar, Ahuran and the centre of Nik Shahr city, 6 samples (1.5%) were found to be infected with Plasmodium vivacs, and 90 samples collected from the Fennoj and Bennett sections of the city had no infection. Samples containing plasmodium falciparum and a mixture of Plasmodium vivacs and Plasmodium falciparum were not detected in this study.Conclusion: The results show that in places where the transmission of both species of Plasmodium vivax and Plasmodium falciparum occur, the detection of malarial parasites by PCR can be a very useful complement to confirm microscopic diagnosis.
